Power Blotter Pre-cut Membranes and Filters, PVDF mini (Invitrogen™)

These Invitrogen Power Blotter Pre-cut Membranes and Filters are designed for transfer of proteins from mini-size gels using the Power Blotter System. The pre-cut membranes and filters include 20 sheets of polyvinylidene difluoride (PVDF) and 80 sheets of absorbent blotting paper (0.85 mm thickness). Power Blotter Pre-cut Membranes and Filters combined with Power Blotter 1-Step Transfer Buffer allow for high-efficiency semi-dry transfer of proteins in less than 10 minutes.

Features of PVDF membranes:
High-quality—PVDF transfer membranes manufactured especially for protein transfer and western blot applications
High-binding affinity—provides excellent protein binding (240 µg/cm2), blocks easily, and is compatible with most organic solvents, acids, and mild bases
Convenient—pre-cut PVDF sheets are ready to use for transfer and blotting experiments from standard precast or homemade mini-gels (8 cm x 8 cm); no need to cut your own membranes, minimizing damage and contamination

The PVDF membrane (0.45 µm pore size, low fluorescence) has higher binding capacity than nitrocellulose. The membrane is compatible with commonly used detection methods such as staining, immunodetection, and fluorescence. The proteins bind to the membrane through hydrophobic interactions. This membrane has lower background levels and increased sensitivity for fluorescent probing than regular PVDF or nitrocellulose membranes.

Thermo Scientific HCT 116 (+hIGF-1) Control Lysate is a stimulated cell lysate that has been validated for the expression levels and phosphorylation of key targets in the RAS signaling pathway.

HCT 116 (+hIGF-1) Control Lysate features include:

The RAS family of GTPase proteins modulates two key signaling pathways involved in cell growth and proliferation: the MAPK and PI3K pathways. Aberrant activation of RAS proteins promotes constitutive activation of cell growth and proliferation that can result in cancer. Mutations in the NRAS, KRAS, and HRAS oncogenes are the most common mutations in human cancer. This lysate has been validated for expression levels of the three isoforms of RAS using mass spectrometry.

The HCT 116 cell line is a human colon cancer epithelial cell line and has been used for proliferation and tumorigenicity studies for colon cancer. The HCT 116 (+hIGF-1) cell lysate is provided as frozen lysate at a standard concentration of 1 mg/mL (500 mg per vial). Lysates are prepared in non-denaturing buffer without loading dyes or dissociating agents (i.e., no 2-mercaptoethanol or dithiothreitol has been added). For western blot analysis, heat lysate to 95°C for 5 minutes. Add reducing agent before heating if desired. The recommended loading volume per lane is 10–20 μL (10–20 µg).

MMP 3-Plex Human ProcartaPlex™ Panel 2 (Invitrogen™)

ProcartaPlex immunoassays are an ideal solution for assessing multiple protein biomarkers in a single sample. Luminex xMAP technology is used for the multi-analyte detection of secreted proteins including cytokines, chemokines, growth factors and other proteins. ProcartaPlex assays are subject to the same stringent and robust development and manufacturing as are our extensively validated Platinum and Instant ELISA kits. We also offer a broad portfolio of pre-configurated panels and combinable single analytes.

Invitrogen Power Blotter 1-Step Transfer Buffer is designed for rapid semi-dry transfer of proteins from polyacrylamide gels (SDS-PAGE) to nitrocellulose or PVDF membranes using the Power Blotter System. Transfer is compatible with commonly used detection methods such as staining, chemiluminescence, or fluorescence.

When used with the Power Blotter System, this buffer provides effective gel-to-membrane transfer of proteins in 5 to 12 minutes with efficiency that is similar to conventional western blotting techniques. Fast blotting methods require a high-current power supply, such as the Power Blotter System, and an optimized high ionic strength transfer buffer, such as Power Blotter 1-Step Transfer Buffer.

By increasing the current, excellent transfer efficiency can be achieved in much shorter time compared to conventional methods. Amperage is held at a constant rate based on the surface area of the transfer stack(s) (~22–23 mA/cm2) and voltage is limited to 25 V.

Power Blotter 1-Step Transfer Buffer is also compatible with other protein semi-dry transfer devices, when they are paired with a suitable high-current power supply. Such devices provide constant high current (1.3 to 5.0 amps) to rapidly transfer proteins via the high ionic strength conditions supplied by the transfer buffer.
