ProcartaPlex immunoassays are an ideal solution for assessing multiple protein biomarkers in a single sample. Luminex xMAP technology is used for the multi-analyte detection of secreted proteins including cytokines, chemokines, growth factors and other proteins. ProcartaPlex assays are subject to the same stringent and robust development and manufacturing as are our extensively validated Platinum and Instant ELISA kits. We also offer a broad portfolio of pre-configurated panels and combinable single analytes.

It is recommended to dilute MMP-2, MMP-3, and MMP-9 samples when measured in serum, heparin plasma,or citrate plasma (EDTA plasma is not recommended). The recommended sample dilution factor is 1:50.

This way, when the basecaller identifies each base and assigns it a quality value, all you have to do is look at the color coding to easily review, discard, or accept it. In addition, the software trims the ends of low-quality bases, grays them out on the user interface for easy identification, and calculates a sample score, which is the average quality value for all the bases in the untrimmed region.

Reduce Data Screening Time
Eliminate manual review of sequencing data batches. With the software's Quality Control (QC) reports, you get read length and sample score (average QV of bases in the clear range) for each sample file, enabling you to sort data by quality. Thermo Scientific BT-549 Control Lysate is a stimulated cell lysate that has been validated for the expression levels and phosphorylation of key targets in the TP53 signaling pathway.

The tumor suppressor protein TP53 functions as a negative checkpoint regulator for abnormal cell signaling, and can induce cell cycle arrest, activate DNA repair proteins, or initiate apoptosis. Mutations in TP53 often result abnormal cell growth and are commonly found in many types of cancer. This lysate has been validated for TP53 expression and phosphorylation.

The BT-549 cell line is a human triple negative epithelial breast cancer cell line derived from mammary gland. The BT-549 cell lysate is provided as frozen lysate at a standard concentration of 1 mg/mL (500 mg per vial). Lysates are prepared in non-denaturing buffer without loading dyes or dissociating agents (i.e., no 2-mercaptoethanol or dithiothreitol has been added). For western blot analysis, heat lysate to 95°C for 5 minutes. Add reducing agent before heating if desired. The recommended loading volume per lane is 10–20 μL (10–20 µg).

The RAS family of GTPase proteins modulates two key signaling pathways involved in cell growth and proliferation: the MAPK and PI3K pathways. Aberrant activation of RAS proteins promotes constitutive activation of cell growth and proliferation that can result in cancer. Mutations in the NRAS, KRAS, and HRAS oncogenes are the most common mutations in human cancer. Ferritin Human ProcartaPlex™ Simplex Kit (Invitrogen™)

The Human Ferritin Simplex ProcartaPlex kit measures ferritin protein and is designed to be combinable with other Simplex kits so that you can create your own multiplex panel that utilizes Luminex xMAP technology for protein detection/quantitation. When combining multiple Simplex kits (i.e., when you are not using a pre-configured Multiplex Panel), only one buffer kit (sold separately) is needed for each assay plate regardless of plex size.

ProcartaPlex immunoassays are based on the principles of a sandwich ELISA, using two highly specific antibodies binding to different epitopes of one protein to quantitate up to 80 protein targets simultaneously when using the FLEXMAP 3D or Luminex 200 instrument and up to 50 protein targets when using the MAGPIX instrument. ProcartaPlex assays require as little as 25 µL of plasma or serum, or 50 µL of cell culture supernatant, and just four hours to obtain analyzed results.

The Luminex MagPlex superparamagnetic microsphere beads in the ProcartaPlex assay are internally dyed with precise proportions of red and infrared fluorophores to create 100 spectrally unique signatures that can be identified by the Luminex xMAP detec¬tion systems (Luminex 200, FLEXMAP 3D, and MAGPIX systems). Similar to a sandwich ELISA, the ProcartaPlex assay uses matched antibody pairs to identify the protein of interest. In a ProcartaPlex multiplex assay, each spectrally unique bead is labeled with antibodies specific for a single target protein, and bound proteins are identified with biotinylated antibodies and streptavidin–R-phycoerythrin (RPE). The conjugation of protein-specific antibodies to a distinct bead allows for analysis of multiple targets in a single well.

The most significant difference between a ProcartaPlex assay and ELISA is that the capture antibody in the ProcartaPlex assay is conjugated to a magnetic bead and not adsorbed to the microplate well, so the ProcartaPlex assay reagents are free-floating in the solution. For detection, the Luminex 200 instrument, for example, contains two lasers, one to distinguish the spectral signature of each bead and the second to quantify the amount of RPE fluorescence, which is proportional to the amount of protein present in the sample. ProcartaPlex multiplex assays can profile up to 80 times more target proteins using significantly less sample in the same time that it takes to perform a traditional sandwich ELISA.
