Variant Reporter™ v3 Upgrade from v2 (Applied Biosystems™)

This is the v2 to v3 upgrade for Variant Reporter Software. This software performs comparative sequencing, also known as direct sequencing, medical sequencing, PCR sequencing, and resequencing with DNA sequencing files. The software is designed for reference-based and non-reference-based analysis such as mutation detection and analysis, SNP discovery and validation, and sequence confirmation. The robust algorithms will call SNPs, mutations, insertions, deletions, and heterozygous insertions⁄deletions for data generated using the Applied Biosystems genetic analyzers.

Note: This product can only be used to upgrade Variant Reporter Software v2 installed on standalone computers (computers separate from the instrument data collection system). Please contact your Sales Representative to upgrade the data collection system and secondary analysis software running on the data collection computer.

TYRO3 Human ProcartaPlex™ Simplex Kit (Invitrogen™)

The Human TYRO-3 Simplex ProcartaPlex kit measures TYRO-3 protein and is designed to be combinable with other Simplex kits so that you can create your own multiplex panel that utilizes Luminex xMAP technology for protein detection/quantitation. When combining multiple Simplex kits (i.e., when you are not using a pre-configured Multiplex Panel), only one buffer kit (sold separately) is needed for each assay plate regardless of plex size.

ProcartaPlex immunoassays are based on the principles of a sandwich ELISA, using two highly specific antibodies binding to different epitopes of one protein to quantitate up to 80 protein targets simultaneously when using the FLEXMAP 3D or Luminex 200 instrument and up to 50 protein targets when using the MAGPIX instrument. ProcartaPlex assays require as little as 25 µL of plasma or serum, or 50 µL of cell culture supernatant, and just four hours to obtain analyzed results.

The Luminex MagPlex superparamagnetic microsphere beads in the ProcartaPlex assay are internally dyed with precise proportions of red and infrared fluorophores to create 100 spectrally unique signatures that can be identified by the Luminex xMAP detec¬tion systems (Luminex 200, FLEXMAP 3D, and MAGPIX systems). Similar to a sandwich ELISA, the ProcartaPlex assay uses matched antibody pairs to identify the protein of interest. In a ProcartaPlex multiplex assay, each spectrally unique bead is labeled with antibodies specific for a single target protein, and bound proteins are identified with biotinylated antibodies and streptavidin–R-phycoerythrin (RPE). The conjugation of protein-specific antibodies to a distinct bead allows for analysis of multiple targets in a single well.

The most significant difference between a ProcartaPlex assay and ELISA is that the capture antibody in the ProcartaPlex assay is conjugated to a magnetic bead and not adsorbed to the microplate well, so the ProcartaPlex assay reagents are free-floating in the solution. For detection, the Luminex 200 instrument, for example, contains two lasers, one to distinguish the spectral signature of each bead and the second to quantify the amount of RPE fluorescence, which is proportional to the amount of protein present in the sample. ProcartaPlex multiplex assays can profile up to 80 times more target proteins using significantly less sample in the same time that it takes to perform a traditional sandwich ELISA.

Thermo Scientific BT-549 Control Lysate is a stimulated cell lysate that has been validated for the expression levels and phosphorylation of key targets in the TP53 signaling pathway.

The tumor suppressor protein TP53 functions as a negative checkpoint regulator for abnormal cell signaling, and can induce cell cycle arrest, activate DNA repair proteins, or initiate apoptosis. Mutations in TP53 often result abnormal cell growth and are commonly found in many types of cancer. This lysate has been validated for TP53 expression and phosphorylation.

The BT-549 cell line is a human triple negative epithelial breast cancer cell line derived from mammary gland. The BT-549 cell lysate is provided as frozen lysate at a standard concentration of 1 mg/mL (500 mg per vial). Lysates are prepared in non-denaturing buffer without loading dyes or dissociating agents (i.e., no 2-mercaptoethanol or dithiothreitol has been added). For western blot analysis, heat lysate to 95°C for 5 minutes. Add reducing agent before heating if desired. The recommended loading volume per lane is 10–20 μL (10–20 µg).

Quantitative proteomic strategies using TMT reagents enable sample multiplexing and precise measurement of protein abundance. However, successful execution of this workflow includes multiple steps that may require optimization, including chromatography, mass spectrometry (MS), and data analysis. A critical parameter for success in this workflow is the ability to consistently detect the TMT fragment ions across the 11 channels of multiplexing. This standard has been specifically designed as a tool to monitor LC-MS/MS system performance for TMT quantitation and MS method optimization and validation.

The Pierce TMT11plex Yeast Digest Standard consists of four congenic strains of Saccharomyces cerevisiae (a parental line, BY4741, and three lines lacking the non-essential proteins Met6, His4, or Ura2) labeled in triplicate (knock-out strains) or duplicate (parental strain) with TMT11-plex label reagents (see figure). The knock-out strains are used in combination with the parental strain to enable comparison of peptide and protein abundance in the deleted and parental strains to monitor system performance for quantitation.

In addition to routine quality control assessment, the standard can also be used as a method development tool to measure the accuracy, precision, and dynamic range of different MS approaches, including optimization of offline fractionation, chromatography, MS acquisition, or data analysis settings.

The multifunctional applications for this standard will allow more reproducible TMT quantitation from day-to-day and experiment-to-experiment.

The AKT signaling pathway serves as a master regulatory pathway for cell growth and proliferation, and phosphorylation plays a major role in its modulation. The A549 cell line is a human lung carcinoma cell line and is used as a model for non-small lung carcinoma. The A549 (+hIGF-1) cell lysate is provided as frozen lysate at a standard concentration of 1 mg/mL (500 mg per vial). Lysates are prepared in non-denaturing buffer without loading dyes or dissociating agents (i.e., no 2-mercaptoethanol or dithiothreitol has been added). For western blot analysis, heat lysate to 95°C for 5 minutes. Add reducing agent before heating if desired. The recommended loading volume per lane is 10–20 μL (10–20 µg).

RAGE Human ProcartaPlex™ Simplex Kit (Invitrogen™)

The Human RAGE Simplex ProcartaPlex kit measures RAGE protein and is designed to be combinable with other Simplex kits so that you can create your own multiplex panel that utilizes Luminex xMAP technology for protein detection/quantitation. When combining multiple Simplex kits (i.e., when you are not using a pre-configured Multiplex Panel), only one buffer kit (sold separately) is needed for each assay plate regardless of plex size.

ProcartaPlex immunoassays are based on the principles of a sandwich ELISA, using two highly specific antibodies binding to different epitopes of one protein to quantitate up to 80 protein targets simultaneously when using the FLEXMAP 3D or Luminex 200 instrument and up to 50 protein targets when using the MAGPIX instrument. For detection, the Luminex 200 instrument, for example, contains two lasers, one to distinguish the spectral signature of each bead and the second to quantify the amount of RPE fluorescence, which is proportional to the amount of protein present in the sample. ProcartaPlex multiplex assays can profile up to 80 times more target proteins using significantly less sample in the same time that it takes to perform a traditional sandwich ELISA.

The RAS family of GTPase proteins modulates two key signaling pathways involved in cell growth and proliferation: the MAPK and PI3K pathways. Aberrant activation of RAS proteins promotes constitutive activation of cell growth and proliferation that can result in cancer. Mutations in the NRAS, KRAS, and HRAS oncogenes are the most common mutations in human cancer. This lysate has been validated for expression levels of the three isoforms of RAS using mass spectrometry.

The HCT 116 cell line is a human colon cancer epithelial cell line and has been used for proliferation and tumorigenicity studies for colon cancer. The HCT 116 (+hIGF-1) cell lysate is provided as frozen lysate at a standard concentration of 1 mg/mL (500 mg per vial). Lysates are prepared in non-denaturing buffer without loading dyes or dissociating agents (i.e., no 2-mercaptoethanol or dithiothreitol has been added). For western blot analysis, heat lysate to 95°C for 5 minutes. Add reducing agent before heating if desired. The recommended loading volume per lane is 10–20 μL (10–20 µg).

The AKT signaling pathway serves as a master regulatory pathway for cell growth and proliferation, and phosphorylation plays a major role in its modulation. Aberrant activation of this pathway promotes survival of tumor cells in many types of cancer. This lysate has been validated for expression of ten key proteins and phosphorylation in the AKT signaling pathway (AKT, PTEN, IRS1, IGF1-R, GSK3a, GSK3b, PRAS40, mTOR, p70S6K, and TSC2).

The MCF-7 cell line is a human epithelial breast cancer cell line with estrogen, progesterone, and glucocorticoid receptor expression and is used as an in vitro model for breast cancer. The MCF7 (+hIGF-1) cell lysate is provided as frozen lysate at a standard concentration of 1 mg/mL (500 mg per vial). Lysates are prepared in non-denaturing buffer without loading dyes or dissociating agents (i.e., no 2-mercaptoethanol or dithiothreitol has been added). For western blot analysis, heat lysate to 95°C for 5 minutes. Add reducing agent before heating if desired. The recommended loading volume per lane is 10–20 μL (10–20 µg).
