Answer

Some customers have isolated RNA from viruses. Unfortunately, the protein coat could cause problems. Another option would be to use the MagMax Viral Isolation Kit.

What kit would you recommend for isolation of miRNA from tissue, cells, or liquid samples? Product FAQ

For tissue or cells, we recommend the mirVana miRNA Isolation Kit (Cat. No. AM1560), while for liquid samples, we recommend the mirVana Paris RNA and Native Protein Purification Kit (Cat. No. AM1556). For high-throughput miRNA isolation we recommend using the MagMAX mirVana Total RNA Isolation kit (Cat. No. A27828)

How should I purify DNA before using the MethylMiner Methylated DNA Enrichment Kit? Product FAQ

Answer

Most commercial available gDNA isolation products can be used and the PureLink Genomic DNA Mini Kit works well. Make sure to work in DNase-free conditions, and that the gDNA remains double-stranded. The MBD2 protein will not bind to ssDNA.

What is the Dynabeads kilobaseBINDER Kit? Product FAQ

The Dynabeads KilobaseBINDER Kit (Cat. No. 60101) is designed for the binding of large (greater than 2 kb) DNA or RNA fragments. The kit contains Dynabeads M-280 Streptavidin magnetic beads plus a unique and patented binding solution. With this binding solution, it is possible to immobilize 70 pmol of a 4 kb DNA fragment and 80 pmol of a 10 kb fragment per mg Dynabeads magnetic beads. This enables various applications to be conducted, including pure template isolation for gene expression, protein-DNA interaction studies, and mRNA processing studies.

What are different methods for cleaning up my miRNA sample? Product FAQ

- Precipitation of miRNA: Follow the instructions for the mirVANA miRNA Isolation Kit or the mirVANA PARIS RNA and Native Protein Purification Kit.
- Double phenol/guanidium (chaotropic reagent) extraction: Disrupt tissue with phenol/guanidine, then extract with phenol, centrifuge, remove the supernatant, and precipitate the RNA.
- Glass-fiber filter: Disrupt the tissue in a chaotropic agent, bind the RNA to a glass-fiber filter, then wash off unbound DNA/protein/salt/nucleotides and elute the RNA.

Can Northern blotting be used to detect small RNAs? Product FAQ

Answer

Yes. You can use a 15% denaturing polyacrylamide gel for Northern analysis of small RNAs, such as miRNAs and siRNAs. A hyridization buffer optimized for use with short probes, such as ULTRAhyb-Oligo solution, should be used for the best results. For more sensitive detection, enrich the RNA sample for small RNAs (e.g., with the mirVana miRNA Isolation Kit or mirVana PARIS RNA and Native Protein Purification Kit. Of course, using a solution hybridization assay such as the mirVana miRNA Detection Kit to analyze small RNAs can provide much greater sensitivity and allow you to detect multiple small RNAs simultaneously.

Which instrument is better for quantitation of purified DNA isolated using the PureLink Microbiome DNA Purification Kit? Product FAQ

Answer

For highly pure samples, both fluorometers (such as the Qubit Flourometric Fluorometer) and spectrophotometers (such as NanoDrop One Spectrophotometer) are successfully utilized and give very similar readings. However, when dealing with DNA or RNA samples that have high levels of contaminants, such as salts or protein, the Qubit Fluorometer provides a more accurate reading, as this instrument uses dyes that are very specific to the analyte of interest. We have seen some challenging microbiome and FFPE samples where the Qubit Fluorometer readings were up to 2 times lower compared to the NanoDrop spectrophotometer, which is overestimating DNA concentrations. More and more laboratories are use both instruments side by side, as they provide complimentary information: the Qubit fluorometer is more accurate and specific in terms of determining nucleic acid concentration, while the NanoDrop spectrophotometer provides a valuable information regarding the purity (ie., A260/280, A260/230).
