The validated dried blood spots (DBS) wash protocol includes centrifugation at 1500 rpm for 30 min. This protocol is suitable for various DBS sample types: desiccated, non-desiccated, >7 days from preparation, ≥7 days from preparation (freshly prepared). A higher rpm is required for DBS samples that have been desiccated for long term storage to adequately remove DNA from the DBS punch.

For short term storage, we recommend storing dried blood spots (DBS) desiccated or non-desiccated at room temperature.
For long term storage, we recommend storing dried blood spots (DBS) at room temperature or at -20 degrees C.

For non-desiccated and freshly prepared samples, 10 min of DBS washing with wash buffer works well, however, customer will need to validate this modified workflow.

The only difference is that the MagMAX CORE Mechanical Lysis Module & Glass Microbeads Bundle with CORE Chemistry (Cat. No. A37488) contains glass microbeads, whereas the other kit (Cat. No. A37487) does not. The beads are also available as a standalone item (Cat. No. A37489) and aid in the homogenization of completely desiccated fecal or environmental samples. These glass microbeads are added to the desiccated sample together with some deionized water, vortexed, and incubated for about 10 minutes prior to the bead beating step.

Can plates be blocked with SuperBlock Blocking Buffer and then stored dry? Product FAQ

Answer

After the plates are blocked, remove the blocking buffer and air dry the plates for several hours. Store plates with desiccant in a plastic bag at 4°C. Change desiccant after 24 hours for optimal storage. The plates should be stable for at least 12 months.

Each HCS CellMask Stain provides sufficient material for labeling approximately 10 x 96-well plates using an assay volume of 100 µL. The product is shipped as two components: a single vial of 250 µg of solid Near-IR Stain (Component A) and a single vial of 100 µL of DMSO (Component B).

We recommend storing Component A at ≤ -20 degrees C, desiccated and protected from light and Component B desiccated at ≤ 25 degrees C.

Answer

For Invitrogen BODIPY 493/503 dye (Cat. No. D3922), one may make a stock solution of 1 mg/mL (3.8 mM) in either absolute ethanol or anhydrous DMSO. After resolubilizing in either solvent, store the stock solution frozen, desiccated, and protected from light. The gradual accumulation of moisture in the solvent will promote aggregation over time, hence store the stock solution desiccated.

For long-term storage, dissolve the dye in absolute ethanol, make smaller aliquots in separate vials, and then evaporate off the ethanol using a vacuum pump. Store the solid dye frozen and protected from light.

Answer

Extracted dried blood spots (DBS) punches obtained from freshly prepared samples appear nearly white/cream. If the DBS sample was prepared >7 days ago or has been desiccated for a prolonged period of time, the color may appear light red/brown. If the extracted DBS punch remains a dark red/brown color, additional wash time may be required.
