Epidermal Growth Factor, Biotinylated, complexed to Alexa Fluor™ 555 Streptavidin (Alexa Fluor™ 555 EGF complex) (Invitrogen™)

We offer several conjugates of epidermal growth factor that are useful for the detection of EGF receptor in cells. Fluorescently labeled EGF has enabled scientists to investigate receptor-membrane interactions, study receptor distribution, calculate rate constants for the interaction of EGF with its receptor, and more. The conjugates incorporating fluorescein (FITC), Oregon Green® 514, and tetramethylrhodamine were constructed with the dye directly attached to EGF. The conjugates of EGF with Alexa Fluor® dyes and Texas Red® dye are complexes of the dye–streptavidin molecule and biotinylated EGF. The biotinylated conjugates utilize biotin-XX, which contains a long spacer arm designed to enhance the probe’s affinity for the EGF receptor.

Epidermal Growth Factor Conjugate Specifications:
• EGF molecule: 53 amino acids, MW=6,045 Da
• Label (Ex/Em): Biotin-XX complexed with Alexa Fluor® 555 streptavidin (~555/565 nm)
• Number of fluorophore molecules on each EGF molecule: ~2–3

TaqMan™ Array Human EGF Pathway (Applied Biosystems™)

The Applied Biosystems® TaqMan® Array Human EGF Pathway 96-well Plate contains 92 assays to EGF pathway associated genes and 4 assays to candidate endogenous control genes. Gene Signature Plates are 96-well plates that are pre-configured with the most appropriate TaqMan® Gene Expression Assays for a specific biological process, pathway, or disease state. Each plate contains predefined assays and endogenous controls dried-down in the wells, ready for accurate assessment of an entire gene signature in one simple experiment.

• Affordable; minimum order of three plates; ideal for small, medium or large-sized projects
• Fast delivery; on your bench in days
• Convenient; dried PCR primer and TaqMan® probe sets supplied in a 96- well plate; just add master mix and your sample
• Same TaqMan®Assay Quality, New Format

TaqMan® Array Plates contain the same high quality TaqMan® Gene Expression Assays (PCR primers and TaqMan® probe sets) that have been commercially available for years in a convenient, dried, 96-well format. Extensive tests have found no difference in performance between TaqMan®Assays supplied in the traditional liquid format and assays dried into a TaqMan® Array Plate. Figure 1 shows the results of an experiment comparing the linear range of the two formats, where no difference in performance was observed.

Just Add Master Mix and Sample, and Begin Cycling
Ideal for projects such as validation of microarray and RNAi data, and pathway studies, the plates include just the right amount of TaqMan®Assay in each well: enough for one 20 µl reaction. You can order as few as three Gene Signature Plates, making these an affordable option for many laboratories and minimizing the waste of unused individually purchased TaqMan®Assays. Save a significant amount of time setting up your experiments. Just add master mix and sample, and begin cycling.

Figure 2 is an example Plate Layout with Gene Symbols and Assay IDs

Plate Map Ensures Proper Data Labeling and Easy Re-Ordering
Each pack of TaqMan® Array Plates is shipped with a CD containing the plate configuration map (see Figure 2) along with the details of each TaqMan®Assay. This information is imported into the real-time PCR instrument to enable proper labeling of experimental data and easy access to Assay ID numbers for simple reordering.

Simple to Use
Each TaqMan® Array Plate well contains a single TaqMan®Assay for one 20 µl reaction. The plates can be stored at room temperature or at 4°C. When you are ready to run your assays, simply add PCR master mix and your cDNA sample, seal the plate, and begin cycling on a real-time PCR instrument.

Compatibility
TaqMan® Array Plates have been optimized for use with TaqMan® Gene Expression Master Mix, but are also compatible with TaqMan® Universal PCR Master Mix. For studies using cultured cells, TaqMan® Array Plates are also compatible (and recommended) with the TaqMan® Gene Expression Cells-to-Ct™ Kit. TaqMan® Array Plates must be run on a real-time PCR instrument with 96-well plate capability. The plates have been validated for use on Applied Biosystems® 7000, 7300, 7500 and 7900HT Fast Real-Time PCR Systems using standard 96-well blocks, standard PCR cycling conditions, and 20 µl reaction

HB-EGF Human ELISA Kit (Invitrogen™)

HB-EGF Human ELISA Kit for ELISA

HB-EGF Human ProcartaPlex™ Simplex Kit (Invitrogen™)

The Human HB-EGF Simplex ProcartaPlex kit measures HB-EGF protein and is designed to be combinable with other Simplex kits so that you can create your own multiplex panel that utilizes Luminex xMAP technology for protein detection/quantitation. When combining multiple Simplex kits (i.e., when you are not using a pre-configured Multiplex Panel), only one buffer kit (sold separately) is needed for each assay plate regardless of plex size.

ProcartaPlex immunoassays are based on the principles of a sandwich ELISA, using two highly specific antibodies binding to different epitopes of one protein to quantitate up to 80 protein targets simultaneously when using the FLEXMAP 3D or Luminex 200 instrument and up to 50 protein targets when using the MAGPIX instrument. ProcartaPlex assays require as little as 25 µL of plasma or serum, or 50 µL of cell culture supernatant, and just four hours to obtain analyzed results.

Flexible panels—design your own panels with Simplex kits to measure your own array of targets
More results per sample—measure up to 80 protein targets in a single 25–50 µL sample
Well-established Luminex technology—the most referenced multiplexing platform for protein detection and quantitation

The Luminex MagPlex superparamagnetic microsphere beads in the ProcartaPlex assay are internally dyed with precise proportions of red and infrared fluorophores to create 100 spectrally unique signatures that can be identified by the Luminex xMAP detec¬tion systems (Luminex 200, FLEXMAP 3D, and MAGPIX systems). Similar to a sandwich ELISA, the ProcartaPlex assay uses matched antibody pairs to identify the protein of interest. In a ProcartaPlex multiplex assay, each spectrally unique bead is labeled with antibodies specific for a single target protein, and bound proteins are identified with biotinylated antibodies and streptavidin–R-phycoerythrin (RPE). The conjugation of protein-specific antibodies to a distinct bead allows for analysis of multiple targets in a single well.

The most significant difference between a ProcartaPlex assay and ELISA is that the capture antibody in the ProcartaPlex assay is conjugated to a magnetic bead and not adsorbed to the microplate well, so the ProcartaPlex assay reagents are free-floating in the solution. For detection, the Luminex 200 instrument, for example, contains two lasers, one to distinguish the spectral signature of each bead and the second to quantify the amount of RPE fluorescence, which is proportional to the amount of protein present in the sample. ProcartaPlex multiplex assays can profile up to 80 times more target proteins using significantly less sample in the same time that it takes to perform a traditional sandwich ELISA.

ProcartaPlex Simplex kits provide the ability to create your own unique panel . More than 90% of ProcartaPlex Simplex targets can be combined, providing you with superior flexibility when creating your own multiplex panel.

ProcartaPlex Simplex kits are available across six species (human, mouse, rat, nonhuman primate, porcine, and canine). Visit thermofisher.com/procartaplex for more information, including a comprehensive list of individual protein targets.
