Simple Viability Determination
SYTOX® Green allows quick determination of cell viability when using flow cytometers, fluorescence microscopes, fluorometers, or fluorescence microplate readers. It will not cross intact membranes, but will easily penetrate compromised membranes characteristic of dead cells. Because it exhibits >500-fold fluorescence enhancement upon binding nucleic acids, briefly incubating dead cells with SYTOX® Green will result in bright green fluorescence with an emission peak of 523 nm when excited by a 488 nm argon-ion laser or any other 450–490 nm source. The exceptional brightness of the signal produced in dead cells makes SYTOX® Green particularly useful for determining viability of not just mammalian cells, but both Gram-positive and Gram-negative bacteria.

SYTOX™ Deep Red Nucleic Acid Stain, for fixed/dead cells (Invitrogen™)

SYTOX Deep Red Nucleic Acid Stain is a high-affinity nucleic acid stain that easily penetrates cells with compromised plasma membranes and yet does not cross the membranes of live cells. With minimal cytoplasmic staining, SYTOX Deep Red stain is particularly useful in immunocytochemistry (ICC), immunohistochemistry (IHC), and immunofluorescence (IF) experiments for staining of nuclei of fixed cells. After a brief incubation with SYTOX Deep Red stain, the nucleic acids of dead or fixed cells fluoresce a bright, deep red/far red and can be detected with a CY5/Deep Red standard filter set or laser configuration.

SYTOX Deep Red Nucleic Acid Stain is multiplexable with blue, green, orange, red, and near IR fluorophores, when compatible fluorescent filter/laser configurations are used. SYTOX Deep Red stain shows increased fluorescence with increasing concentrations of dsDNA, but does not show much affinity for RNA or ssDNA. It has a bright initial signal and excellent photo stability in a typical imaging experiment. These properties make SYTOX Deep Red stain a simple and quantitative single-step dead/fixed cell nucleus-labeling dye for use with fluorescence micro¬scopes, fluorimeters, fluorescence microplate readers, and flow cytometers.

SYTOX Deep Red Nucleic Acid Stain had been used successfully in mono-layer cells, thin-sliced tissue, thick-sliced tissue, 3D cell cultures/spheroids, and bacteria. It is supplied at a 1 mM concentration in DMSO (~2000X concentration). A concentration of 0.5 µM (1X) is recommended for most applications and cell types. In fluorescent imaging of various mammalian cells, a concentration range of 2.5 µM to 0.25 µM has been used. For viability testing by flow cytometry, a concentration of 1 µM to 10 nM has been used for Jurkat cells and a range of 4 µM to 0.5 µM has been used with E. coli.

SYTO™ Deep Red Nucleic Acid Stain, for live cells (Invitrogen™)

SYTO Deep Red Nucleic Acid Stain is cell-permeant dye that specifically stains the nuclei of live, dead, or fixed cells. It can be used in live-cell fluorescent imaging workflows and in fixed cell workflows, including immunocytochemistry, immunohistochemistry, and immunofluorescence experiments. After a brief incubation with the stain, the nuclei of live, dead, or fixed cells fluoresce with a deep red/far red signal that is detectable with a Cy5/deep red standard filter set or laser configuration.

SYTO Deep Red Nucleic Acid Stain can be mulitplexed with blue, green, orange, red, and near-IR fluorophores when compatible fluorescent filter/laser configurations are used. The stain increases in fluorescence with increasing concentrations of dsDNA. SYTO Deep Red Nucleic Acid Stain displays up to five times more fluorescence when incubated with dsDNA vs RNA, unlike DRAQ or other live-cell deep red nucleic acid stains. NucRed Dead 647 ReadyProbes Reagent is a cell impermeant stain that emits bright far-red fluorescence when bound to DNA. Since this reagent stains the cells without plasma membrane integrity it is extremely useful to measure viability and to measure cytotoxicity in kinetic live cell assays. NucRed Dead 647 reagent is also suitable for staining nuclei in fixed cell preparations and tissue sections.

NucGreen Dead 488 ReadyProbes Reagent is a room temperature-stable solution of SYTOX Green Nucleic Acid Stain (Cat. No. S7020), which is a cell-impermeant stain that emits exceptionally bright, green fluorescence when bound to DNA. Cells that have lost plasma membrane integrity are stained within minutes, making this an extremely useful stain to estimate live/dead cell ratios and to measure cytotoxicity in kinetic live-cell assays. It is also suitable for staining nuclei in fixed cell preparations and tissue sections.

Cell imaging applications
The membrane-impermeable NucGreen Dead 488 reagent is a high-affinity DNA stain that easily stains cells with compromised cell membranes, yet does not enter living cells. It is therefore ideal for discrimination of live and dead cells. The fluorescence increases more than 500-fold on binding to dsDNA in cells with compromised cell membranes (dying or dead cells), in fixed cells, and tissue slices. The spectral properties, with excitation/emission at 504/523 nm when bound to DNA, are ideal for standard FITC filters. .

Suggestions for use
• NucGreen Dead 488 reagent may be added directly to cells in full media or buffer solutions
• It easily stains nuclei of fixed cells and tissue slices
• In most cases, 2 drops/ml and an incubation time of 15 to 30 minutes will give bright nuclear staining; however, optimization may be needed for some cell types, conditions, and applications. In such cases, simply add more or fewer drops until the optimal staining intensity is obtained. In most cases, staining intensity increases with time if cells are not washed prior to imaging.
• NucGreen Dead 488 dye is excited with a maximum at 504 nm when bound to DNA, with an emission maximum at 523 nm. It is detected through standard GFP and FITC filters.

Rapid and convenient, with a large dynamic range
The CyQUANT® Direct assay is a fluorescence-based proliferation and cytotoxicity assay for microplate readers with a linear detection range from less than 100 to 20,000 cells per well in most cell types. The no-wash, homogenous format and fast add-mix-read protocol makes the CyQUANT® Direct assay ideal for HTS applications. The assay can be completed in one hour, with no washes, no cell lysis, or temperature equilibrations required. The signal is stable for several hours, affording additional work flow convenience.

How it works
The CyQUANT® Direct assay is based on a cell-permeant DNA-binding dye in combination with a masking dye reagent. Cellular DNA is highly regulated and estimates of cell numbers based on DNA are very accurate. The masking dye blocks staining of dead cells or cells with compromised cell membranes resulting in only healthy cells being stained. The CyQUANT® Direct assay, therefore, measures proliferation as well as membrane integrity, another measure of cell health. Users of metabolically based cell health assays can readily compare historical data with results from the CyQUANT® Direct assay, as concordance assays based on metabolism or energy (ATP) has been shown to be excellent. Furthermore, it is possible to multiplex with spectrally distinct fluorescent probes or luminescence assay kits because cells are not lysed. Furthermore, the fluorescent signal allows the user to switch between HTS and HCS plate readers if multiplex assays require different platforms.

iPrep™ Trizol™ Plus RNA Kit (Invitrogen™)

Use the iPrep™ Trizol® Plus RNA Kit for the highest purity, even from difficult samples such as fatty tissue. This kit provides reproducible RNA sample preparation from up to 50 mg of tissue, 10e7 cultured cells, 10 ml blood, and plant samples. The kit contains pre-filled, fully qualified cartridges to avoid any mix-ups or cross-contamination. It also provides low CVs and reduced purification variability, increasing the reliability of your gene expression assays.

PrepFiler™ Forensic DNA Extraction Kit, updated (Applied Biosystems™)

The PrepFiler Forensic DNA Extraction Kit uses magnetic particles with an optimized multi-component surface chemistry to deliver superior DNA yield from most forensic sample types, including body fluids, and stains and swabs of body fluids. Whether it’s an automated or manual application, this kit is designed to help improve the overall yield and concentration of DNA recovered from both routine and challenging samples, while enabling removal of PCR inhibitors. This means greater quantities of highly purified DNA for improved downstream genotyping results. This kit is intended for manual use with 1.5 mL tubes. Improved Packaging and Configuration
Several packaging and configuration improvements have been implemented for the updated PrepFiler kits:
• An additional wash buffer, PrepFiler Wash Buffer B, has now been incorporated into the kits to minimize the potential for detergent carryover from PrepFiler Wash Buffer A, which can inhibit downstream PCR applications when the maximum volume of DNA extract is used for subsequent PCR amplification.
• The kit packaging has been updated to include all reagents, including magnetic particles, in the same kit box, with a single expiration date.
• Shipping will occur at ambient temperature, and the kits can also be stored upon receipt at ambient temperature.

mRNA Catcher™ PLUS Purification Kit (Invitrogen™)

The mRNA Catcher™ PLUS Kit enables high-throughput purification of mRNA from cells, tissues, blood, and total RNA in a 96-well format. By taking advantage of Locked Nucleic Acid (LNA) technology, the mRNA Catcher™ PLUS Kit provides improved performance and a faster protocol, while preserving mRNA yield and purity (see table). The mRNA Catcher™ PLUS Kit offers:

Robust performance—high binding capacity per well (>80 ng/well)
Reproducible results—low CVs from well-to-well and plate-to-plate (~2%)

How it works
mRNA from total RNA, cells, tissue, and blood samples is hybridized to an immobilized, single-stranded 20-mer oligo(dT)-containing LNA. The incorporation of LNA into the oligo probe increases the hybridization efficiency and specificity of the mRNA binding. The mRNA can either be eluted or directly reversed-transcribed to cDNA directly in the well. The mRNA Catcher™ PLUS method does not require centrifugation, precipitation, phase separation, or column filtration. In addition, the entire method is amenable to automation for streamlined, high-throughput expression profiling for large-scale processes. Isolated mRNA is suitable for use in downstream applications such as RT-PCR and qRT-PCR.

FluoReporter™ Blue Fluorometric dsDNA Quantitation Kit, 200-2,000 assays (Invitrogen™)

The FluoReporter® Blue Fluorometric dsDNA Quantitation Kit provides the protocols and reagents for analyzing cellular NA with the blue-fluorescent Hoechst 33258 nucleic acid stain. With this kit, quantitation of cellular DNA is rapid, and all manipulations can be carried out in microplate wells. The cells are lysed by freezing them in distilled water, the diluted dye solution is then added to the lysed cells, and fluorescence is measured.
