This PIK3CA E542K/PIK3R1 (p110 alpha E542K/p85 alpha) recombinant human protein (full length) was expressed in insect cells. PIK3CA E542K/PIK3R1 (p110 alpha E542K/p85 alpha) lipid kinase is comprised of a 110 kDa catalytic subunit and an 85 kDa regulatory subunit. A number of isoforms of the 110 kDa catalytic subunit and the 85 kDa regulatory subunit exist in cells. PIK3CA is frequently mutated or amplified in a variety of cancers including ovarian and colon and this kinase is one of the PI3K mutants. PIK3CA gene copy number is increased in over 30% of ovarian cancers and this leads to increased PI3-kinase activity. Furthermore, the activity of PIK3CA is essential for vascular development and inactivation of PIK3CA leads to severe defects in angiogenic sprouting and vascular remodeling. Phosphatidylinositol 3-kinase (PI3K) is composed of a catalytic subunit (p110δ) and a regulatory subunit (p85α in this case), which are encoded by the PIK3CD and PIK3R1 genes. PI3K phosphorylates phosphoinositides on the 3-hydroxyl group of the inositol ring. It plays a role in oncogenic transformation and is consistently expressed at a high level in AML blast cells. PI3K can play a selective role in the amplification of phosphatidylinositol (3,4,5) trisphosphate (PIP3) levels that leads to neutrophil polarization, and regulates directional migration, but not random locomotion. The PI3K signaling pathway is involved in immune responses relevant to the pathogenesis of rheumatoid arthritis and other inflammatory diseases.

PIK3CB/PIK3R1 (p110β/p85α) is a phosphoinositide-3-kinase (PI3K) comprised of a catalytic subunit, p110β, and a regulatory subunit, p85α, encoded by the PIK3CB and PIK3R1 genes respectively. As with other members of the PI3K family, p110β has important regulatory roles in cellular processes including cell survival, proliferation, and differentiation. It affects growth factor signaling and survival by phosphorylating inositol lipids. PTEN-deficient cancer cells have been observed to depend on p110β signaling.
