Recombinant Human Transforming Growth Factor β 1 (TGFB1) is a bioactive protein intended for use in cell culture applications. TGFB1 is a multifunctional peptide that controls proliferation, differentiation, and other functions in many cell types. TGFB1 acts synergistically with TGF-α in inducing transformation. It also acts as a negative autocrine growth factor. Dysregulation of TGFB1 activation and signaling may result in apoptosis.

Optimal concentration for individual applications should be determined by a dose response assay.

ACVR2B is a type II member of the TGF-beta family of receptor ser⁄thr kinases and is involved in the activin and myostatin signaling pathway.

BMPR1A (ALK3) Recombinant Human Protein

BMPR1A is a Type I member of the TGF beta receptor superfamily of transmembrane ser⁄thr kinases which phosphorylates intracellular SMADs in response to bone morphogenic proteins.

ACVR1 (ALK2) [R206H] Recombinant Human Protein

ACVR1 (ALK2) belongs to the transforming growth factor-beta (TGF-beta) superfamily of structurally related signaling proteins which encodes activin A type I receptor. ACVR1 signals a particular transcriptional response in concert with activin type II receptors. Mutations in ACVR1 are associated with fibrodysplasia ossificans progressiva (FOP) and the mutations are located in or adjacent to the GS regulatory region or active site of the kinase. The mutations in ACVR1 were predicted by protein structure homology modeling to activate the ACVR1 protein and enhance receptor signaling. Dysregulated ACVR1 kinase activity plays a role in the pathogenesis of FOP. Recombinant human ACVR1 (R206H) (147-end) was expressed by baculovirus in Sf9 insect cells using an N-terminal GST tag.

Noggin Recombinant Human Protein (Gibco™)

Recombinant Human Noggin is a bioactive protein intended for use in cell culture applications. Noggin binds and inactivates members of the transforming growth factor-beta (TGF-β) superfamily signaling proteins, such as bone morphogenetic protein-4 (BMP4). By diffusing through extracellular matrices more efficiently than members of the TGF-β superfamily, this protein may have a principal role in creating morphogenic gradients. The protein appears to have pleiotropic effect, both early in development as well as in later stages. It was originally isolated from Xenopus based on its ability to restore normal dorsal-ventral body axis in embryos that had been artificially ventralized by UV treatment. The results of the mouse knockout of the ortholog suggest that it is involved in numerous developmental processes, such as neural tube fusion and joint formation.

Recombinant TGFA is a bioactive protein intended for use in cell culture applications. TGFA shows about 40% sequence homology with epidermal growth factor and competes with EGF for binding to the EGF receptor, stimulating its phosphorylation and producing a mitogenic response.

The LanthaScreen® Eu Kinase Binding Assay is a TR-FRET assay that lets you measure inhibitor binding to non-activated kinases, perform simple off-rate measurements, and makes simple the study of type III (allosteric) inhibitors.

The LanthaScreen® Eu Kinase Binding Assay is easy to optimize and run. The assay uses a simple mix-and-read format, with a single one hour incubation and no development steps. Binding of an Alexa Fluor® conjugate or “tracer’ to a tagged kinase is detected by addition of a europium-labeled anti-tag antibody, in this case anti-GST. Simultaneous binding of the tracer and antibody to the kinase results in a high degree of fluorescence resonance energy transfer (FRET), while displacement of the tracer with a kinase inhibitor results in a loss of FRET (Fig 1).
