The Porcine TNF-alpha Simplex ProcartaPlex kit measures TNF-alpha protein and is designed to be combinable with other Simplex kits so that you can create your own multiplex panel that utilizes Luminex xMAP technology for protein detection/quantitation. When combining multiple Simplex kits (i.e., when you are not using a pre-configured Multiplex Panel), only one buffer kit (sold separately) is needed for each assay plate regardless of plex size.

ProcartaPlex immunoassays are based on the principles of a sandwich ELISA, using two highly specific antibodies binding to different epitopes of one protein to quantitate up to 80 protein targets simultaneously when using the FLEXMAP 3D or Luminex 200 instrument and up to 50 protein targets when using the MAGPIX instrument. ProcartaPlex assays require as little as 25 µL of plasma or serum, or 50 µL of cell culture supernatant, and just four hours to obtain analyzed results.

The Luminex MagPlex superparamagnetic microsphere beads in the ProcartaPlex assay are internally dyed with precise proportions of red and infrared fluorophores to create 100 spectrally unique signatures that can be identified by the Luminex xMAP detec¬tion systems (Luminex 200, FLEXMAP 3D, and MAGPIX systems). Similar to a sandwich ELISA, the ProcartaPlex assay uses matched antibody pairs to identify the protein of interest. In a ProcartaPlex multiplex assay, each spectrally unique bead is labeled with antibodies specific for a single target protein, and bound proteins are identified with biotinylated antibodies and streptavidin–R-phycoerythrin (RPE). The conjugation of protein-specific antibodies to a distinct bead allows for analysis of multiple targets in a single well.

The most significant difference between a ProcartaPlex assay and ELISA is that the capture antibody in the ProcartaPlex assay is conjugated to a magnetic bead and not adsorbed to the microplate well, so the ProcartaPlex assay reagents are free-floating in the solution. For detection, the Luminex 200 instrument, for example, contains two lasers, one to distinguish the spectral signature of each bead and the second to quantify the amount of RPE fluorescence, which is proportional to the amount of protein present in the sample. ProcartaPlex multiplex assays can profile up to 80 times more target proteins using significantly less sample in the same time that it takes to perform a traditional sandwich ELISA.

Tumor necrosis factor alpha (TNF-α) recombinant human protein is supplied as a lyophilized powder. This protein is suitable for use in analysis of protein structure and performing cell based assays. It can also be used as an immunogen, as a protein standard, or in other research applications.

This recombinant protein is expressed from a DNA sequence encoding the soluble form of human TNF (NP_000585.2) (Val 77-Leu 233) retaining the initial Met at the N-terminus.

N-terminal Sequence Analysis: Met.

Activity: measured in a cytotoxicity assay using L929 mouse fibrosarcoma cells in the presence of the metabolic inhibitor actinomycin D. The EC50 for this effect is typically 25-200 pg/mL, corresponding to a specific activity of 5 x 106 to 4 x 107 units/ mg.

Formulation: lyophilized in 140 mM NaCl, 2.7 mM KCl, 10 mM Na2HPO4, 1.8 mM KH2PO4, pH 7.4, 5% mannitol, 5% trehalose, 0.02% Tween®-80.

Reconstitution: Dissolve the protein in sterile double-distilled water to a concentration of 0.2 mg/mL or lower. It is recommended that the protein be aliquoted and used as soon as possible. Store aliquots under sterile conditions at -20°C. Avoid repeated freeze-thaw cycles.

Expiration Date: expires one year from date of receipt when stored as instructed.

Background
Tumor necrosis factor alpha (TNF-α), also known as cachectin and TNFSF1A, is the prototypic cytokine of the TNF superfamily, and is a multifunctional molecule involved in the regulation of a wide spectrum of biological processes including cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. TNF-α is produced mainly by macrophages, and large amounts of this cytokine are released in response to lipopolysaccharide, other bacterial products, and Interleukin-1 (IL-1). TNF-α is primarily produced as a type I I transmembrane protein arranged in stable homotrimers, and a soluble trimeric form of TNF-α is released via proteolytic cleavage by the metalloprotease TACE/ ADAM17. Two receptors mediating the TNF-α function have been identified, the ubiquitous TNF RI from which most signaling are derived, and the hematopoietic cell-restricted TNF RII. Upon the TNF-α trimers binding, TNF receptors also form trimers, undergo a conformational change, and enable the adaptor protein TRADD to bind to the death domain, and thus initiates the signal pathways such as the NF-kB, Jak/STAT, and the MAPK pathways, as well as the death signaling. As a major mediator of apoptosis, inflammation and immunity, TNF-α has been implicated in the pathogenesis of a variety of human diseases including autoimmune diseases, insulin resistance, and cancer.
