What is the main purpose of TMT (Tandem Mass Tag) reagents? Product FAQ

Answer

TMT technology is used to perform multiplex quantitation of proteins extracted from cells and tissues. Rather than quantify single proteins (for example with an antibody), here, large numbers of proteins can be quantified in a single run.

What is the main purpose of Tandem Mass Tag (TMT) reagents? Product FAQ

Answer

Tandem Mass Tag technology is used to individually label different protein samples so they can be combined into a single sample for LC-MS analysis. The major advantages of this workflow are higher sample throughput, less instrument analysis time, high precision of peptide quantitation among replicates, and fewer missing quantified proteins among different samples.

What is the difference between Relative and Absolute Quantitation? Which type of quantitation is used for proteomic samples? Product FAQ

Answer

Relative quantitation compares the amount of an analyte in different samples. Absolute quantitation measures the actual amount of an analyte in a sample using a known standard at different concentrations. Label-free, SILAC, and TMT (Tandem Mass Tag) workflows are used for relative quantitation of protein samples. Heavy (i.e., AQUA) peptides are used as standard for absolute quantitation of target proteins.

After running the Pierce TMT11plex Yeast Digest Standard, I observe low signal to noise ratio and poor quantitation accuracy for the Tandem Mass Tag (TMT) reporter ions. What do you recommend? Product FAQ

Answer

Here are some recommendations:

- Clean and recalibrate the mass spectrometry instrument using one of our Pierce Calibration Solutions (https://www.thermofisher.com/search/browse/category/us/en/600654).
- Verify settings for liquid chromatography (LC) acquisition methods.
- Fractionate TMT-labeled samples using the Pierce High pH Reversed-Phase Peptide Fractionation Kit (Cat. No. 84868) to reduce sample complexity.
