Epidermal Growth Factor, Tetramethylrhodamine Conjugate (rhodamine EGF) (Invitrogen™)

We offer several conjugates of epidermal growth factor that are useful for the detection of EGF receptor in cells. Fluorescently labeled EGF has enabled scientists to investigate receptor-membrane interactions, study receptor distribution, calculate rate constants for the interaction of EGF with its receptor, and more. The conjugates incorporating fluorescein (FITC), Oregon Green® 514, and tetramethylrhodamine were constructed with the dye directly attached to EGF. The conjugates of EGF with Alexa Fluor® dyes and Texas Red® dye are complexes of the dye–streptavidin molecule and biotinylated EGF. The biotinylated conjugates utilize biotin-XX, which contains a long spacer arm designed to enhance the probe’s affinity for the EGF receptor.

Epidermal Growth Factor Conjugate Specifications:
• EGF molecule: 53 amino acids, MW=6,045 Da
• Label (Ex/Em): Tetramethylrhodamine (~555/580 nm)
• Number of fluorophore molecules on each EGF molecule: 1
• Fluorescence is typically monitored using a flow cytometer, fluorescence microscope, or fluorimeter

Cytokine/Chemokine/Growth Factor 45-Plex Human ProcartaPlex™ Panel 1 (Invitrogen™)

The Human Cytokine & Chemokine & Growth Factor 45-plex ProcartaPlex Panel is a preconfigured multiplex immunoassay kit that measures 45 protein targets using Luminex xMAP technology. ProcartaPlex preconfigured panels are supplied with all the necessary reagents to perform the assay and are validated to ensure reproducibility and consistency. These panels are designed for those who need greater validation for longitudinal studies.

ProcartaPlex immunoassays are based on the principles of a sandwich ELISA, using two highly specific antibodies binding to different epitopes of one protein to quantitate all protein targets simultaneously using a Luminex instrument. ProcartaPlex multiplex assays require as little as 25 µL of plasma or serum, or 50 µL of cell culture supernatant, and just four hours to obtain analyzed results.

Reproducible, reliable results—validated as a panel to the highest industry standard, including protein target combinability and cross-reactivity testing
More results per sample—measure multiple protein targets simultaneously in a single 25–50 µL sample
Well-established Luminex technology—the most referenced multiplexing platform for protein detection and quantitation

ProcartaPlex assays utilize Luminex xMAP (multianalyte profiling) technology for the simultaneous detection and quantitation of up to 80 protein targets in a single 25–50 µL sample—from plasma, serum, cell culture supernatants, and other bodily fluids.

The Luminex MagPlex superparamagnetic microsphere beads in the ProcartaPlex assay are internally dyed with precise proportions of red and infrared fluorophores to create 100 spectrally unique signatures that can be identified by the Luminex xMAP detec¬tion systems (Luminex 200, FLEXMAP 3D, and MAGPIX systems). Similar to a sandwich ELISA, the ProcartaPlex assay uses matched antibody pairs to identify the protein of interest. In a multiplexed assay, each spectrally unique bead is labeled with antibodies specific for a single target protein, and bound proteins are identified with biotinylated antibodies and streptavidin–R-phycoerythrin (RPE). The conjugation of protein-specific antibodies to a distinct bead allows for analysis of multiple targets in a single well.

The most significant difference between a ProcartaPlex assay and ELISA is that the capture antibody in the ProcartaPlex assay is conjugated to a magnetic bead and not adsorbed to the microplate well, so the ProcartaPlex assay reagents are free-floating in the solution. For detection, the Luminex 200 instrument, for example, contains two lasers, one to distinguish the spectral signature of each bead and the second to quantify the amount of RPE fluorescence, which is proportional to the amount of protein present in the sample. ProcartaPlex multiplex assays can profile more target proteins using significantly less sample in the same time that it takes to perform a traditional sandwich ELISA.

ProcartaPlex multiplex panels are available in multiple formats across six species (human, mouse, rat, nonhuman primate, porcine, and canine). Visit thermofisher.com/procartaplex for more information and available products.

Geltrex™ LDEV-Free, hESC-Qualified, Reduced Growth Factor Basement Membrane Matrix (Gibco™)

Geltrex™ LDEV-Free hESC-qualified Reduced Growth Factor Basement Membrane Matrix is a soluble form of basement membrane extracted from murine Engelbreth-Holm-Swarm (EHS) tumors and is free of viruses, including lactose dehydrogenase elevating virus (LDEV), sometimes found in this type of preparation. Routinely used for attachment and maintenance of human embryonic stem cells (hESC), every lot of Geltrex™ LDEV-free hESC-qualified Reduced Growth Factor Basement Membrane Matrix is quality control tested using hESCs and StemPro hESC Serum Free Media to help ensure growth and pluripotency of each lot.

Key Features of Geltrex™ LDEV-Free hESC-qualified Reduced Growth Factor Basement Membrane Matrix:

• Every lot is qualified using hESC, which helps reduce the need for customers to lot screen
• Free of the lactose dehydrogenase elevating virus (LDEV) making it safe for all types of cell culture
• Consistent protein concentration lot-to-lot helps ensure reproducible results

Gain Reliability and Consistency in Your hESC Studies
Human ESCs have specific needs for attachment and maintenance and some lots of basement membrane extract may not provide adequate support. Geltrex™ hESC-qualified reduced growth factor basement membrane matrix has been tested for support of hESC growth and pluripotency, therefore minimizing the need for you to test multiple lots to find one that works. Our consistent manufacturing process helps ensure a protein concentration averaging 15 mg/ml with very little variation from lot-to-lot, so you can have confidence in every purchase.

Free of LDEV Contamination
Recent advances in processing EHS tumor cells have resulted in virus free basement membrane extracts. Every lot is tested by tested by RT-PCR methods so you can now have confidence that every lot of Geltrex™ is virus-free.
