ProcartaPlex assays utilize Luminex xMAP (multianalyte profiling) technology for the simultaneous detection and quantitation of up to 80 protein targets in a single 25–50 µL sample—from plasma, serum, cell culture supernatants, and other bodily fluids.

The Luminex MagPlex superparamagnetic microsphere beads in the ProcartaPlex assay are internally dyed with precise proportions of red and infrared fluorophores to create 100 spectrally unique signatures that can be identified by the Luminex xMAP detec¬tion systems (Luminex 200, FLEXMAP 3D, and MAGPIX systems). Similar to a sandwich ELISA, the ProcartaPlex assay uses matched antibody pairs to identify the protein of interest. In a multiplexed assay, each spectrally unique bead is labeled with antibodies specific for a single target protein, and bound proteins are identified with biotinylated antibodies and streptavidin–R-phycoerythrin (RPE). The conjugation of protein-specific antibodies to a distinct bead allows for analysis of multiple targets in a single well.

The most significant difference between a ProcartaPlex assay and ELISA is that the capture antibody in the ProcartaPlex assay is conjugated to a magnetic bead and not adsorbed to the microplate well, so the ProcartaPlex assay reagents are free-floating in the solution. For detection, the Luminex 200 instrument, for example, contains two lasers, one to distinguish the spectral signature of each bead and the second to quantify the amount of RPE fluorescence, which is proportional to the amount of protein present in the sample. ProcartaPlex multiplex assays can profile more target proteins using significantly less sample in the same time that it takes to perform a traditional sandwich ELISA.

Epidermal Growth Factor, Biotin-XX Conjugate (biotin EGF) (Invitrogen™)

This purified epidermal growth factor (EGF) contains a single biotin-XX molecule on the N-terminal amino acid. Biotin-XX EGF contains a long spacer arm that enhances the probe’s affinity for the EGF receptor and facilitates binding of a label such as a dye–streptavidin molecule, a Qdot® nanocrystals–streptavidin molecule, or an enzyme-conjugated streptavidin. Epidermal Growth Factor, Biotinylated, complexed to Alexa Fluor™ 488 Streptavidin (Alexa Fluor™ 488 EGF complex) (Invitrogen™)

We offer several conjugates of epidermal growth factor that are useful for the detection of EGF receptor in cells. Fluorescently labeled EGF has enabled scientists to investigate receptor-membrane interactions, study receptor distribution, calculate rate constants for the interaction of EGF with its receptor, and more. The conjugates incorporating fluorescein (FITC), Oregon Green® 514, and tetramethylrhodamine were constructed with the dye directly attached to EGF. The conjugates of EGF with Alexa Fluor® dyes and Texas Red® dye are complexes of the dye–streptavidin molecule and biotinylated EGF. The biotinylated conjugates utilize biotin-XX, which contains a long spacer arm designed to enhance the probe’s affinity for the EGF receptor.

• EGF molecule: 53 amino acids, MW=6,045 Da
• Label (Ex/Em): Biotin-XX complexed with Alexa Fluor® 488 streptavidin (~495/519 nm)

pHrodo™ Green Epidermal Growth Factor (EGF) Conjugate (Invitrogen™)

pHrodo® Green Epidermal Growth Factor (EGF) Conjugate is used for the detection of internalized EGF receptor in cells. Fluorescently labeled EGF has enabled scientists to investigate receptor-membrane interactions, study receptor distribution, calculate rate constants for the interaction of EGF with its receptor, and more. Fluorescent labels that respond to a pH change add another level of detection. pHrodo® Green is weakly fluorescent outside of cells at neutral pH, but when internalized specifically through EGF endocytosis, it becomes brightly fluorescent in the acidic endosomes. This allows the measurement of internalized EGF, as well as the dynamic monitoring of EGF internalization in live cells.
