This recombinant protein was expressed from a DNA sequence encoding the extracellular domain (Met 1-Lys 642) of human EGFR (NP_005219) fused to a polyhistidine tag at the C-terminus.

Activity: Measured by its binding ability in a functional ELISA. Immobilized human EGF at 10 µg/ml can bind human EGFR with a linear range of 3.2-400 ng/ml.

Formulation: Lyophilized in 20 mM Tris, pH7.4, 50 mM NaCl, 3 mM DTT, 0.5 mM TCEP, 5% Mannitol, 5% Trehalose, 0.02% Tween-80.

Reconstitution: Dissolve the protein in sterile double distilled water to a concentration of 0.2 mg/ml or lower. It is recommended that the protein be aliquoted and be used as soon as possible. Store aliquots under sterile conditions at -20°C. Avoid repeated freeze-thaw cycles.

Met proto-oncogene/hepatocyte growth factor receptor (MET/c-MET), recombinant human protein is supplied as a lyophilized powder. It is suitable for use in protein studies such as protein structure analysis and protein-protein interactions. In general, recombinant proteins can also be used as an immunogen, as a protein standard, or in cell biology research applications.

This recombinant protein was expressed from a DNA sequence encoding the extracellular domain (Met 1-Thr 932) of human MET (NP_000236) fused to a polyhistidine tag at the C-terminus.

Activity: Measured by its binding ability in a functional ELISA. Immobilized recombinant human HGF at 10 µg/ml (100 µl/well) can bind biotinylated MET. The ED50 of biotinylated Met is 5.28 µg/ml.

This recombinant protein is expressed from a DNA sequence encoding the soluble form of human TNF (NP_000585.2) (Val 77-Leu 233) retaining the initial Met at the N-terminus.

N-terminal Sequence Analysis: Met.

Activity: measured in a cytotoxicity assay using L929 mouse fibrosarcoma cells in the presence of the metabolic inhibitor actinomycin D. The EC50 for this effect is typically 25-200 pg/mL, corresponding to a specific activity of 5 x 106 to 4 x 107 units/ mg.

Background
Tumor necrosis factor alpha (TNF-α), also known as cachectin and TNFSF1A, is the prototypic cytokine of the TNF superfamily, and is a multifunctional molecule involved in the regulation of a wide spectrum of biological processes including cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. TNF-α is produced mainly by macrophages, and large amounts of this cytokine are released in response to lipopolysaccharide, other bacterial products, and Interleukin-1 (IL-1). TNF-α is primarily produced as a type I I transmembrane protein arranged in stable homotrimers, and a soluble trimeric form of TNF-α is released via proteolytic cleavage by the metalloprotease TACE/ ADAM17. Two receptors mediating the TNF-α function have been identified, the ubiquitous TNF RI from which most signaling are derived, and the hematopoietic cell-restricted TNF RII. Upon the TNF-α trimers binding, TNF receptors also form trimers, undergo a conformational change, and enable the adaptor protein TRADD to bind to the death domain, and thus initiates the signal pathways such as the NF-kB, Jak/STAT, and the MAPK pathways, as well as the death signaling. Background
Human epidermal growth factor (EGF) exerts actions on a wide variety of biological processes, and plays an important role in the regulation of cell growth, proliferation, and differentiation mediated by its high-affinity receptor EGFR on the cell surface. It induces differentiation of specific cells, promotes angiogenesis, and inhibits gastric acid secretion in vivo, while serves as a potent mitogenic actor for a variety of cultured cells of both ectodermal and mesodermal origin. Human EGF was originally isolated from urine based on its inhibitory effect on gastric secretion and named urogastrone accordingly. EGF is the founding member of the EGF-family of proteins which are characterized by the presence of at least one EGF structural unit (a conserved 6 cysteine motif that forms three disulfide bonds). The EGF precursor is believed to exist as a membrane-bound molecule containing 9 EGF units, and is proteolyticallycleaved to generate the 53-amino acid peptide hormone.
