pHrodo™ Red Epidermal Growth Factor (EGF) Conjugate (Invitrogen™)

pHrodo® Red Epidermal Growth Factor (EGF) Conjugate is used for the detection of internalized EGF receptor in cells. Fluorescently labeled EGF has enabled scientists to investigate receptor-membrane interactions, study receptor distribution, calculate rate constants for the interaction of EGF with its receptor, and more. Fluorescent labels that respond to a pH change add another level of detection. pHrodo® Red is weakly fluorescent outside of cells at neutral pH, but when internalized specifically through EGF endocytosis, it becomes brightly fluorescent in the acidic endosomes. This allows the measurement of internalized EGF, as well as the dynamic monitoring of EGF internalization in live cells.

Cytokine/Chemokine/Growth Factor 37-Plex NHP ProcartaPlex™ Panel (Invitrogen™)

The Non-Human Primate Cytokine & Chemokine & Growth Factor 37-plex ProcartaPlex Panel is a preconfigured multiplex immunoassay kit that measures 37 protein targets using Luminex xMAP technology. ProcartaPlex preconfigured panels are supplied with all the necessary reagents to perform the assay and are validated to ensure reproducibility and consistency. These panels are designed for those who need greater validation for longitudinal studies.

ProcartaPlex assays utilize Luminex xMAP (multianalyte profiling) technology for the simultaneous detection and quantitation of up to 80 protein targets in a single 25–50 µL sample—from plasma, serum, cell culture supernatants, and other bodily fluids.

The Luminex MagPlex superparamagnetic microsphere beads in the ProcartaPlex assay are internally dyed with precise proportions of red and infrared fluorophores to create 100 spectrally unique signatures that can be identified by the Luminex xMAP detec¬tion systems (Luminex 200, FLEXMAP 3D, and MAGPIX systems). Similar to a sandwich ELISA, the ProcartaPlex assay uses matched antibody pairs to identify the protein of interest. In a multiplexed assay, each spectrally unique bead is labeled with antibodies specific for a single target protein, and bound proteins are identified with biotinylated antibodies and streptavidin–R-phycoerythrin (RPE). The conjugation of protein-specific antibodies to a distinct bead allows for analysis of multiple targets in a single well.

The most significant difference between a ProcartaPlex assay and ELISA is that the capture antibody in the ProcartaPlex assay is conjugated to a magnetic bead and not adsorbed to the microplate well, so the ProcartaPlex assay reagents are free-floating in the solution. For detection, the Luminex 200 instrument, for example, contains two lasers, one to distinguish the spectral signature of each bead and the second to quantify the amount of RPE fluorescence, which is proportional to the amount of protein present in the sample. ProcartaPlex multiplex assays can profile more target proteins using significantly less sample in the same time that it takes to perform a traditional sandwich ELISA.

Geltrex™ LDEV-Free Reduced Growth Factor Basement Membrane Matrix (Gibco™)

Geltrex™ LDEV-Free Reduced Growth Factor Basement Membrane Matrix is a soluble form of basement membrane extracted from murine Engelbreth-Holm-Swarm (EHS) tumors and is free of viruses, including lactose dehydrogenase elevating virus (LDEV), sometimes found in this type of preparation. The major components of Geltrex™ matrix include laminin, collagen IV, entactin, and heparin sulfate proteoglycans. It is ideal for the promotion and maintenance of differentiated phenotypes in a variety of cell cultures including primary epithelial cells, endothelial cells, smooth muscle cells, and human induced pluripotent stem cells (iPSC). pHrodo™ Green Epidermal Growth Factor (EGF) Conjugate (Invitrogen™)

pHrodo® Green Epidermal Growth Factor (EGF) Conjugate is used for the detection of internalized EGF receptor in cells. Fluorescently labeled EGF has enabled scientists to investigate receptor-membrane interactions, study receptor distribution, calculate rate constants for the interaction of EGF with its receptor, and more. Fluorescent labels that respond to a pH change add another level of detection. pHrodo® Green is weakly fluorescent outside of cells at neutral pH, but when internalized specifically through EGF endocytosis, it becomes brightly fluorescent in the acidic endosomes. This allows the measurement of internalized EGF, as well as the dynamic monitoring of EGF internalization in live cells.

ProcartaPlex assays utilize Luminex xMAP (multianalyte profiling) technology for the simultaneous detection and quantitation of up to 80 protein targets in a single 25–50 µL sample—from plasma, serum, cell culture supernatants, and other bodily fluids.

The Luminex MagPlex superparamagnetic microsphere beads in the ProcartaPlex assay are internally dyed with precise proportions of red and infrared fluorophores to create 100 spectrally unique signatures that can be identified by the Luminex xMAP detec¬tion systems (Luminex 200, FLEXMAP 3D, and MAGPIX systems). Similar to a sandwich ELISA, the ProcartaPlex assay uses matched antibody pairs to identify the protein of interest. In a multiplexed assay, each spectrally unique bead is labeled with antibodies specific for a single target protein, and bound proteins are identified with biotinylated antibodies and streptavidin–R-phycoerythrin (RPE). The conjugation of protein-specific antibodies to a distinct bead allows for analysis of multiple targets in a single well.

The most significant difference between a ProcartaPlex assay and ELISA is that the capture antibody in the ProcartaPlex assay is conjugated to a magnetic bead and not adsorbed to the microplate well, so the ProcartaPlex assay reagents are free-floating in the solution. For detection, the Luminex 200 instrument, for example, contains two lasers, one to distinguish the spectral signature of each bead and the second to quantify the amount of RPE fluorescence, which is proportional to the amount of protein present in the sample. ProcartaPlex multiplex assays can profile more target proteins using significantly less sample in the same time that it takes to perform a traditional sandwich ELISA.

Plate Map Ensures Proper Data Labeling and Easy Re-Ordering
Each pack of TaqMan® Array Plates is shipped with a CD containing the plate configuration map (see Figure 2) along with the details of each TaqMan®Assay. This information is imported into the real-time PCR instrument to enable proper labeling of experimental data and easy access to Assay ID numbers for simple reordering.

Simple to Use
Each TaqMan® Array Plate well contains a single TaqMan®Assay for one 20 µl reaction. The plates can be stored at room temperature or at 4°C. When you are ready to run your assays, simply add PCR master mix and your cDNA sample, seal the plate, and begin cycling on a real-time PCR instrument.

Compatibility
TaqMan® Array Plates have been optimized for use with TaqMan® Gene Expression Master Mix, but are also compatible with TaqMan® Universal PCR Master Mix. Epidermal growth factor receptor (EGFR), recombinant human protein is supplied as a lyophilized powder. It is suitable for use in studying protein-protein interactions, and cell biology research applications. It can also be used as a protein standard.

Since this product is a truncated protein and does not contain intracellular catalytic activity domain, it is not suitable for use in enzyme activity studies.

This recombinant protein was expressed from a DNA sequence encoding the extracellular domain (Met 1-Gly 645) of human EGFR (NP_005219) fused to the Fc region of human IgG1 at the C-terminus.

Activity: Measured by its binding ability in a functional ELISA. Immobilized recombinant human EGF at 10 µg/ml (100 µl/well) can bind human EGFR with a linear range of 0.64-400 ng/ml.

Formulation: Lyophilized in PBS, pH7.4, 5% Mannitol, 5% Trehalose, and 0.02% Tween-80.

Reconstitution: Dissolve the protein in sterile double distilled water to a concentration of 0.2 mg/ml or lower. It is recommended that the protein be aliquoted and be used as soon as possible. Store aliquots under sterile conditions at -20°C. Avoid repeated freeze-thaw cycles.

Expiration Date: Expires one year from date of receipt when stored as instructed.
