Soluble TNF-RII Recombinant Human Protein Gibco™

Recombinant Soluble TNF-RII is a bioactive protein intended for use in cell culture applications. sTNF-RII is a member of the TNF-receptor superfamily. This protein and TNF-receptor 1 form a heterocomplex that mediates the recruitment of two anti-apoptotic proteins, c-IAP1 and c-IAP2, which possess E3 ubiquitin ligase activity. The function of IAPs in TNF-receptor signalling is unknown, however, c-IAP1 is thought to potentiate TNF-induced apoptosis by the ubiquitination and degradation of TNF-receptor-associated factor 2, which mediates anti-apoptotic signals. Knockout studies in mice also suggest a role of this protein in protecting neurons from apoptosis by stimulating antioxidative pathways.

Background
Tumor necrosis factor alpha (TNF-α), also known as cachectin and TNFSF1A, is the prototypic cytokine of the TNF superfamily, and is a multifunctional molecule involved in the regulation of a wide spectrum of biological processes including cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. TNF-α is produced mainly by macrophages, and large amounts of this cytokine are released in response to lipopolysaccharide, other bacterial products, and Interleukin-1 (IL-1). TNF-α is primarily produced as a type I I transmembrane protein arranged in stable homotrimers, and a soluble trimeric form of TNF-α is released via proteolytic cleavage by the metalloprotease TACE/ ADAM17. Two receptors mediating the TNF-α function have been identified, the ubiquitous TNF RI from which most signaling are derived, and the hematopoietic cell-restricted TNF RII. Upon the TNF-α trimers binding, TNF receptors also form trimers, undergo a conformational change, and enable the adaptor protein TRADD to bind to the death domain, and thus initiates the signal pathways such as the NF-kB, Jak/STAT, and the MAPK pathways, as well as the death signaling. As a major mediator of apoptosis, inflammation and immunity, TNF-α has been implicated in the pathogenesis of a variety of human diseases including autoimmune diseases, insulin resistance, and cancer.

IL4 Recombinant Swine Protein Gibco™

Recombinant IL4 is a bioactive protein intended for use in cell culture applications. IL4 promotes the proliferation and differentiation of activated B cells. In addition, IL4 up-regulates class II MHC antigen expression and IgE receptors.

IP10 Recombinant Mouse Protein Gibco™

Recombinant IP10 is a bioactive protein intended for use in cell culture applications. IP10 is a chemokine of the CXC subfamily and ligand for the receptor CXCR3. Binding of this protein to CXCR3 results in pleiotropic effects, including stimulation of monocytes, natural killer and T-cell migration, and modulation of adhesion molecule expression.

Leptin Recombinant Human Protein Gibco™

Recombinant Leptin is a bioactive protein intended for use in cell culture applications. Leptin acts through the leptin receptor and functions as part of a signaling pathway that can inhibit food intake and/or regulate energy expenditure to maintain constancy of the adipose mass. This protein also has several endocrine functions, and is involved in the regulation of immune and inflammatory responses, hematopoiesis, angiogenesis and wound healing.

TNF (TNFA) Recombinant Cynomolgus Protein

Tumor Necrosis Factor (TNF), recombinant cynomolgus protein is supplied as a lyophilized powder. It is suitable for use in analysis of protein structure, and performing cell based assays. It can also be used as an immunogen, as a protein standard, or in other research applications

This recombinant protein was expressed from a DNA sequence encoding the soluble form of cynomolgus TNF-alpha (Val 77-Leu 233) fused to an initial Met at the N-terminus.

Activity: Measured in a cytotoxicity assay using L-929 mouse fibrosarcoma cells in the presence of the metabolic inhibitor actinomycin D. The ED50 is typically 20-80 pg/ml.

Formulation: Lyophilized in 50 mM Tris, 50 mM NaCl, pH 7.4, 5% Mannitol, 5% Trehalose, 0.02% Tween 80.

Reconstitution: Dissolve the protein in sterile double distilled water to a concentration of 0.2 mg/ml or lower. It is recommended that the protein be aliquoted and be used as soon as possible. Store aliquots under sterile conditions at -20°C. Avoid repeated freeze-thaw cycles.

Expiration Date: Expires one year from date of receipt when stored as instructed.

BMP4 Recombinant Human Protein Gibco™

Recombinant Human Bone Morphogenetic Protein 4 (BMP4)is a bioactive protein intended for use in cell culture applications. BMP4 is a member of the bone morphogenetic protein family which is part of the transforming growth factor-beta superfamily. The superfamily includes large families of growth and differentiation factors. Bone morphogenetic proteins were originally identified by an ability of demineralized bone extract to induce endochondral osteogenesis in vivo in an extraskeletal site. This particular family member plays an important role in the onset of endochondral bone formation in humans.
