ProcartaPlex assays utilize Luminex xMAP (multianalyte profiling) technology for the simultaneous detection and quantitation of up to 80 protein targets in a single 25–50 µL sample—from plasma, serum, cell culture supernatants, and other bodily fluids.

The Luminex MagPlex superparamagnetic microsphere beads in the ProcartaPlex assay are internally dyed with precise proportions of red and infrared fluorophores to create 100 spectrally unique signatures that can be identified by the Luminex xMAP detec¬tion systems (Luminex 200, FLEXMAP 3D, and MAGPIX systems). Similar to a sandwich ELISA, the ProcartaPlex assay uses matched antibody pairs to identify the protein of interest. In a multiplexed assay, each spectrally unique bead is labeled with antibodies specific for a single target protein, and bound proteins are identified with biotinylated antibodies and streptavidin–R-phycoerythrin (RPE). The conjugation of protein-specific antibodies to a distinct bead allows for analysis of multiple targets in a single well.

The most significant difference between a ProcartaPlex assay and ELISA is that the capture antibody in the ProcartaPlex assay is conjugated to a magnetic bead and not adsorbed to the microplate well, so the ProcartaPlex assay reagents are free-floating in the solution. For detection, the Luminex 200 instrument, for example, contains two lasers, one to distinguish the spectral signature of each bead and the second to quantify the amount of RPE fluorescence, which is proportional to the amount of protein present in the sample. ProcartaPlex multiplex assays can profile more target proteins using significantly less sample in the same time that it takes to perform a traditional sandwich ELISA.

IL23 Recombinant Human Protein (Gibco™)

Recombinant Human IL23 is a bioactive protein intended for use in cell culture applications. IL23 is important in the inflammatory response. IL23 induces the secretion of IFN-γ, and stimulates the proliferation of Human T cells. Recombinant Human IL23 is one of the many Gibco® recombinant bioactive proteins available from Life Technologies. This is the 100 microgram size.

IL13 Recombinant Human Protein

Interleukin 13 (IL13), recombinant human protein is supplied as a lyophilized powder. It is suitable for use in analysis of protein structure, and performing cell based assays. It can also be used as an immunogen, as a protein standard, or in other research applications

This recombinant protein was expressed from a DNA sequence encoding human IL-13 (AAK53823.1) (Met 1 - Asn 132).

Activity: Measured in a cell proliferation assay using TF1 human erythroleukemic cells (Kitamura, T. et al. 1989, J. Cell Physiol. 140:323). The ED50 for this effect is typically 0.75-3.5 ng/ml.

Formulation: Lyophilized in 140 mM NaCl, 2.7 mM KCl, 10 mM Na2HPO4, 1.8 mM KH2PO4, pH 7.4, 5% Mannitol, 5% Trehalose, 0.02% Tween 80.

Reconstitution: Dissolve the protein in sterile double distilled water to a concentration of 0.2 mg/ml or lower. It is recommended that the protein be aliquoted and be used as soon as possible. Store aliquots under sterile conditions at -20°C. Avoid repeated freeze-thaw cycles.

Expiration Date: Expires one year from date of receipt when stored as instructed.

Recombinant IFN-γ is a bioactive protein intended for use in cell culture applications. Recombinant rat IFN-γ is intended for use in cell culture applications. IFN-γ is involved in many biological processes such as inducing the secretion of TNF-α and stimulating the release of reactive oxygen species from macrophages. It is also involved with bone growth and inhibits bone reesorption. It is antiviral and antiparasitic and inhibits the proliferation of cells.

TPO is a humoral growth factor that is necessary for megakaryocyte proliferation and maturation, as well as for thrombopoiesis.
