The Biotin-Binding Plate Sample Pack helps in assay development. Thermo Scientific Pierce Copper Coated High Binding Capacity (HBC) Plates use an exclusive coating process to increase the amount of histidine-tagged protein that will bind to the plate surface.

Features of Copper Coated High Binding Capacity Plates:

• Wider dynamic range with four-fold greater capacity than regular nickel chelate-coated plate
• Copper chelate provides greater binding capacity than nickel
• The detection limit is 0.1 pg of polyhistidine tagged fusion protein per well

These copper coated plates are ideal for analyzing polyhistidine-tagged fusion proteins by ELISA-based methods. Proteins that contain a succession of several histidine residues at the amino or carboxyl terminus have a strong binding affinity for metals such as copper. Pierce Copper Coated Plates have high binding capacity because they are coating using an exclusive process that increases the amount of histidine-tagged protein that will bind to the plate surface. Although copper is less discriminating in its binding specificity than Nickel, the higher binding capacity of these plates is ideal for high throughput screening applications that need improved sensitivity and greater dynamic range. The copper-coated plates are available as clear, white or black plates that can be used with colorimetric, chemiluminescent or fluorescent detection methods, respectively.

Note: Copper coated plates are for use with purified His-tagged proteins. Typical cell lysates contain non-his-tagged proteins that may bind to copper, which can reduce binding capacity and signal-to-noise ratios. For cell lysates, use Thermo Scientific Pierce Nickel Coated Plates.

Thermo Scientific NeutrAvidin Protein Coated Black 384-Well Plates are pre-blocked and ready-to-use polystyrene plates for binding biotinylated antibodies or probes for ELISA and other target-specific assays.

Features of NeutrAvidin Protein Coated Black 384-Well Plates:

• Black plates for fluorescent detection methods

• Easy and gentle immobilization of biotin-containing conjugates
• Low nonspecific binding
• No denaturing of the protein component of a conjugate upon binding

Biotin-binding plates are ideal for applications when direct adsorption to polystyrene denatures antibodies or the target molecule. Streptavidin is the most popular and widely available biotin-binding protein for research methods. Streptavidin has an isoelectric point of 5 to 6, resulting in low nonspecific interactions. Thermo Scientific NeutrAvidin Protein is a deglycosylated form of avidin, which has a near-neutral pI (6.3), resulting in even lower nonspecific interactions. NeutrAvidin Protein has the additional advantage of lacking the bacterial RYD sequence found on streptavidin, which eliminates off-target binding to the RGD binding domain of adhesion receptors present in a variety of cells.
