Pierce™ Copper Coated High Capacity Plates, Black, 96-Well Thermo Scientific™

Thermo Scientific Pierce Copper Coated High Binding Capacity (HBC) Plates use an exclusive coating process to increase the amount of histidine-tagged protein that will bind to the plate surface.

These copper coated plates are ideal for analyzing polyhistidine-tagged fusion proteins by ELISA-based methods. Proteins that contain a succession of several histidine residues at the amino or carboxyl terminus have a strong binding affinity for metals such as copper. Pierce Copper Coated Plates have high binding capacity because they are coating using an exclusive process that increases the amount of histidine-tagged protein that will bind to the plate surface. Although copper is less discriminating in its binding specificity than Nickel, the higher binding capacity of these plates is ideal for high throughput screening applications that need improved sensitivity and greater dynamic range. The copper-coated plates are available as clear, white or black plates that can be used with colorimetric, chemiluminescent or fluorescent detection methods, respectively.

Features of Copper Coated High Binding Capacity Plates:

• Quantitate previously undetectable histidine-tagged proteins
• Wider dynamic range with four-fold greater capacity than regular nickel chelate-coated plate
• Copper chelate provides greater binding capacity than nickel

Note: Copper coated plates are for use with purified His-tagged proteins. Typical cell lysates contain non-his-tagged proteins that may bind to copper, which can reduce binding capacity and signal-to-noise ratios. For cell lysates, use Thermo Scientific Pierce Nickel Coated Plates.

These Thermo Scientific Pierce Maleimide Activated Black, 96-Well Plates are ideal for binding sulfhydryl-containing molecules that are difficult to coat onto polystyrene plates, such as peptides that contain a terminal cysteine. Our coated plates are an especially useful tool for assessing specific anti-hapten antibody titers during antibody production.

Features of Maleimide Activated Black, 96-Well Plates:

• Black, for fluorescent assays
• Pre-blocked to reduce nonspecific binding
• Easy (spontaneous) immobilization of peptides derivatized with a terminal cysteine and proteins with free sulfhydryl
• Binding Capacity: 100 to 150 pmol of a sulfhydryl-containing peptide (307 Da) per well
• Activation Level: 100 µL

Maleimides react with free sulfhydryl group(s) at pH of 6.5-7.5, forming stable thioether linkages. Reaction with amines becomes significant at pH > 7.5. Some sulfhydryl-containing peptides and proteins may oxidize in solution and form disulfide bonds, which cannot react with maleimides. Disulfide bonds can be reduced to produce free sulfhydryls. The TCEP Disulfide Reducing Gel enables peptide or protein reduction while recovering the sample in the absence of reducing agents. Sulfhydryls can be introduced via amine modification using N-succinimidyl S-acetylthioacetate (SATA) or 2-iminothiolane-HCl (Traut's Reagent).

Pierce™ Protein A Coated Plates, White, 96-Well Thermo Scientific™

Thermo Scientific Pierce Protein A Coated Plates provide an alternative to direct, passive adsorption methods for immobilizing antibodies for ELISA and other plate-based assay techniques.

These plates are uniformly and stably coated with Protein A, and enable antibodies or pre-established antibody-antigen complexes to be captured to wells of the polystyrene plate for subsequent detection in plate readers. The strategy is useful for a variety of assay situations, but it is generally not suitable for sandwich-type ELISA.

Features of the plates:

• Retain antibody activity, which can be lost when antibodies are immobilized by passive adsorption
• Orient antibodies for maximum antigen-binding capacity
• Immobilize antibodies for plate assays without prior purification
• Ensure minimal variation (<5% well-to-well) from consistent coating
• Reduce nonspecific binding because plates are pre-blocked with SuperBlock Blocking Buffer

These Thermo Scientific Pierce Maleic Anhydride Activated 8-Well Strip Plates are clear, 96-well strip plates that allow attachment of amine-containing peptides and other molecules to microplate wells for use in binding assays.

Features of Maleic Anhydride Activated Plates:

These plates are useful for immobilizing peptides and other ligands that do not coat efficiently by passive adsorption. Reaction of the maleic anhydride groups with primary amines (-NH2) results in formation of amide bonds that are stable at neutral pH and above. Acidic conditions hydrolyze the bond; for example, the half-life of hydrolysis at pH 3.5 and 37°C is approximately 11 hours (Ref.1). Therefore, coating reactions are best performed at pH 8-9, and the reacted plates are best used at pH > 7 for ELISA and other methods.
