MagMAX™-96 Viral RNA Isolation Kit (Applied Biosystems™)

The MagMAX-96 Viral RNA Isolation Kit is for the isolation of viral RNA and DNA from diverse sample types (e.g., environmental samples and biofluids). Benefits of MagMAX magnetic bead-based purification:

With this kit, DNA and RNA input levels ranging from 20 copies to 25 x 106 copies have been quantitatively recovered from plasma, serum, and milk. And even with less sample input, pathogen detection was as sensitive (equivalent CTs) or more sensitive (lower CTs) using nucleic acid isolated with the MagMAX kit when compared to nucleic acid isolated using another company's product.

This kit is optimized for automated nucleic acid purification using KingFisher magnetic particle processors. This kit can also be used for manual nucleic acid purification using a 96-well magnetic ring stand. The Magnetic Stand-96 (Cat. No. AM10027) and the 96-well Magnetic Ring Stand (Cat. No. AM10050) are designed for paramagnetic bead precipitation from standard 96-well, U-bottom microplates. The Magnetic-Ring Stand pellets the magnetic beads in a circle shape, enabling easy removal of supernatant by a robot or multichannel pipettors.

Benefits of the MagMAX™ Total Nucleic Acid Isolation Kit:
The MagMAX™ Total Nucleic Acid Isolation Kit procedure combines an effective method sample disruption with magnetic bead–based nucleic acid isolation and purification technology. The MagMAX™ microspherical paramagnetic beads have a large available binding surface and can be fully dispersed in solution, allowing thorough nucleic acid binding, washing, and elution. This results in consistent yields of high-quality nucleic acid with >50% recovery and little sample-to-sample variation (recovery may vary depending on sample type). The entire procedure can be completed in approximately 1 hour.

Overview of the Procedure:
Samples are disrupted using zirconia beads in a guanidinium thiocyanate–based solution; this rapidly releases nucleic acid while simultaneously inactivating nucleases. Samples are then diluted with isopropanol, and paramagnetic beads are added to the sample. The beads bind nucleic acids and are then immobilized on magnets and washed to remove proteins and other contaminants. A second wash solution is used to remove residual binding solution. Finally, nucleic acid is eluted in a small volume of low-salt buffer. The MagMAX™ Total Nucleic Acid Isolation Kit procedure is designed for processing samples in 96-well plates, thus 96 samples can be processed at once; however, the kit can also be used with fewer than 96 samples.

The sensitivity of viral RNA detection was compared using RNA isolated with Applied Biosystems® MagMAX™ protocol or the current National Veterinary Services Laboratories (NVSL) validated viral testing protocol. Even at one-tenth the sample input, viral RNA isolated using the MagMAX™ kit could be detected at equal or greater levels than RNA isolated using the NVSL protocol and recommended competitor's kit (see data). The MagMAX™-96 Total RNA Isolation Kit is designed for the rapid, moderate-throughput purification of RNA from animal and plant cells and tissue. RNA purified using the kit is ideal for PCR and RT-PCR applications. The kit uses magnetic bead–based purification and includes sufficient reagents for 96 reactions. Unlocking the information contained in FFPE samples is essential to cancer research. Common nucleic acid isolation methods require you to choose which nucleic acid to recover from a given sample, with loss of the other nucleic acid (and any information that may have resulted from its analysis). Alternatively, the DNA and RNA are co-purified, or the sample is split in half so that each can be purified.

More information from less sample input
The MagMAX FFPE DNA/RNA Ultra Kit protocol addresses these issues by first isolating DNA from the FFPE sample and then recovering the RNA from the DNA-depleted supernatant. The nucleic acids are prepared as separate eluates and are then ready for downstream analysis. This allows for a fuller analysis of precious samples, including investigation of important biomarkers, such as hotspot mutations, CNVs, gene fusions, and indels.

Benefits of MagMAX magnetic bead–based purification
The magnetic bead–based purification format of the kit allows for easy scaling of the number of samples processed either manually with a magnetic stand or with a magnetic particle processor such as the KingFisher™ Duo Prime or KingFisher™ Flex processor.

The MagMAX Viral/Pathogen Ultra Nucleic Acid Isolation Kit contains the same reagents as the standard MagMAX Viral/Pathogen Nucleic Acid Isolation Kit (Cat. No. A42352) and so, like that kit, recovers nucleic acid from viral particles and gram negative bacteria. However, the MagMAX Viral/Pathogen Ultra kit also includes a proprietary enzyme mix that enables lysis and nucleic acid recovery from the toughest pathogens, such as gram-positive bacteria, yeast, and fungi. MagMAX™ DNA Multi-Sample Kit (Invitrogen™)

MagMAX™ DNA Multi-Sample Kits are designed for rapid purification of high-quality genomic DNA from a variety of sample types including blood and blood cards, tissue, mouse tails, buccal swabs, buffy coats, and cultured cells. These purification kits use MagMAX™ magnetic bead-based nucleic acid isolation technology to produce high yields of purified DNA, free from inhibitors that may affect downstream reactions such as genotyping assays, CE and next-generation sequencing, and high resolution melting analysis. The 50 prep kit is designed for manual microfuge tube processing.

The demands for DNA yield and purity have become more stringent with the development of new analytical methods and technologies, such as high-throughput genotyping, and next-generation DNA sequencing platforms. MagMAX™ DNA Multi-Sample Kits meet the challenge by delivering highly pure DNA that is free of nucleases, proteins, and other inhibitors of downstream enzymatic reactions.
As shown in Table 1, DNA with high yield and purity can be obtained from a variety of materials using the MagMAX™ DNA Multi-Sample Kits. This is in contrast to filter membrane–based methods that often require different kit configurations for various input materials. For applications requiring maximal yields, for small batch sizes, or for DNA archival purposes MagMAX™ DNA Multi-Sample Kit (tube format, manual processing) may be desirable because larger sample inputs are accommodated. DNA yields suitable for most routine applications may be obtained using the MagMAX™-96 DNA Multi-Sample Kits (96-well plate format, compatible with automated handling) (Table 1).

The MagMAX™ DNA Multi-Sample Kit also delivers high yields of DNA from lipid or glycogen rich samples, such as brain and liver tissues, respectively. Figure 1 shows more than twice as much DNA was purified from tissues with the MagMAX™ DNA Multi-Sample Kit than with the leading competitor’s filter membrane–based method.

Most enzymatic molecular applications are prone to inhibition by analytes carried over from the sample matrix (e.g., heme, heparin, salts, protein) or the purification method itself (phenol, salts, alcohol). Figure 2 illustrates the effect of DNA purity on sensitive molecular applications, such as quantitative PCR (qPCR). DNA was extracted from blood using the MagMAX™-96 DNA Multi-Sample Kit, or a competitor’s filter membrane–based kit and subjected to real-time PCR analysis. The cycle threshold CT values obtained with the DNA purified using the MagMAX™ DNA Multi-Sample Kit were lower than those obtained with the filter membrane kit–purified DNA, indicating the MagMAX™ kit yields higher-purity DNA.

MagMAX™ DNA Multi-Sample Kits provide purified DNA that is suitable for a variety of Applied Biosystems application workflows (Figure 3), including SNP genotyping and DNA sequencing analysis (see Figure 4).
