PowerUp™ SYBR™ Green Master Mix (Applied Biosystems™)

For qPCR users using SYBR green dye who need to minimize optimization time, Applied Biosystems PowerUp SYBR Green Master Mix offers proprietary Dual-Lock hot start technology. Unlike other polymerases, a Dual-Lock polymerase offers two unique hot start mechanisms to prevent primer dimer and non-specific amplification.

PowerUp SYBR Green Master Mix is a pre-formulated, optimized, universal 2X master mix for real-time PCR workflows. Coupled with user-supplied primer sets and template, PowerUp SYBR Green Master Mix is designed to amplifiy targets for accurate gene expression analysis.

Exceptional specificity with dual hot-start mechanism
Specificity is paramount in obtaining high-quality data in SYBR Green reactions and can be enhanced through control of the Taq DNA polymerase at lower temperatures before stringent primer binding. PowerUp SYBR Green Master Mix uses the Dual-Lock Taq DNA polymerase enzyme that combines two unique hot-start mechanisms to help control its activity and to prevent undesirable early activity of the polymerase at low temperatures. In an experiment evaluating 24 different primer sets, PowerUp SYBR Green Master Mix generated single melt curves 100% of the time, consistent with highly specific amplification.

Tight reproducibility in CTs over a wide dynamic range
PowerUp SYBR Green Master Mix demonstrates excellent reproducibility over a wide dynamic range for a variety of targets tested (see figure). In addition, this master mix provides efficient amplification over 6 logs of sample input. Unlike competitor mixes that can exhibit inhibition at high cDNA inputs and low correlation coefficients over a dynamic range experiment, PowerUp SYBR Green Master Mix efficiency is maintained over the full dynamic range to help deliver maximal confidence in data quality (see figure).

UDG included for carryover contamination control1
The inclusion of uracil-DNA glycosylase (UDG) and dUTP in the PowerUp SYBR Green Master Mix enables the degradation of any previously amplified PCR products, helping prevent contamination of subsequent qPCR reactions and possible false positives.

Stability of pre-assembled reactions
The tight control of the Dual-Lock Taq DNA polymerase in PowerUp SYBR Green Master Mix enables reactions to be assembled up to 72 hours prior to cycling without impacting data (see figure). This consistency allows users to have confidence and flexibility in the use of PowerUP SYBR Green Master Mix across numerous workflow scenarios.2

Broad instrument compatibility
PowerUp SYBR Green Master Mix can be used in either standard or fast cycling mode and is compatible with all Applied Biosystems real-time PCR instruments.3 It is also compatible with the Bio-Rad IQ™5 and CFX96™/CFX384™ systems, Roche LightCycler™ LC480, and Stratagene™ MX3005P™ systems. TaqMan Fast Virus 1-Step Master Mix is designed for fast, highly sensitive real-time RT-PCR even in the presence of challenging PCR inhibitors. High sensitivity
Many common research samples with viral nucleic acids have very low levels of target. We have optimized the master mix for high-sensitivity detection of viral targets. The higher-concentration master mix allows you to set up smaller reactions, which means that you can perform fast cycling protocols and obtain at least the same sensitivity as is expected from standard-cycling qPCR. Alternatively, larger sample input amounts can be added to standard reaction volumes for more accurate quantification of low-titer samples (Figure 1).

Consistent results in the presence of inhibitors
Research samples commonly assayed for viruses include blood, dirt, and tissues. Buffer components and proprietary additives in the TaqMan Fast Virus 1-Step Master Mix have been optimized to handle RT-PCR inhibitors (Figure 2) to help ensure consistent performance even with these difficult samples, so that you can be more confident in your results.

Flexibility in targets
It is common for labs to test for both RNA and DNA viruses in a variety of samples. To simplify your experiments, a single TaqMan Fast Virus 1-Step Master Mix protocol has been developed to assay both types of nucleic acid, so you can perform RNA and DNA virus queries next to each other on the same plate using the same handling steps (Figure 3). Additionally, because virus research often includes multiplexed primers and probes and internal reaction controls, we have optimized the master mix to work with multiple target amplicons (Figure 4).

Fast results
The TaqMan Fast Virus 1-Step Master Mix speeds your time to results and maximizes the use of your real-time PCR instruments with a fast protocol. The high-concentration formulation allows for more target nucleic acid sample to be added in the smaller reaction volumes required to run fast protocols. This enables you to maintain sensitivity with low-titer research samples while improving speed and throughput (Figure 5).

High accuracy genotyping with purified or crude lysate sample inputs
Accurate genotyping is important when limited sample is available or PCR inhibitors are unavoidable, such as in clinical diagnostic testing. TaqPath ProAmp Master Mix has been optimized to yield accurate and reproducible results, even in low-volume reactions prepared with purified or crude lysate sample inputs. The figure below shows a comparison of genotyping accuracy versus competitor mixes for the same set of TaqMan genotyping assays using crude lysate from blood samples. TaqPath ProAmp Master Mix has been optimized to provide high-specificity allelic discrimination as demonstrated by excellent cluster separation in genotyping plots, whether the sample is purified genomic DNA or crude lysate.

High accuracy copy number quantitation
The versatile TaqPath ProAmp Master Mix can also be used for highly accurate determination of copy number variation using purified genomic DNA samples. The figure below shows a comparison of copy number variation results using TaqPath ProAmp Master Mix versus two other mixes. RNA Quantification Kit for SYBR Green I and ROX™ Passive Reference Dye (Applied Biosystems™)

Accurate quantification from limited RNA concentrations

Target preparation assay kits such as GeneChip™ WT Pico Kit and GeneChip 3' IVT Pico Kit allow expression profiling using as little as 100 pg of total RNA (approximately 10 cells). These low amounts of RNA for such target preparation assays require accurate measurement of the quantity of input sample RNA for optimal performance. However, at these low RNA amounts, it becomes virtually impossible to accurately quantify the RNA with standard UV absorbance or fluorescent dye–based techniques.

The RNA quantification kit uses Power SYBR Green RNA-to-CT 1-Step Kit to measure levels of the consistently expressed 18S ribosomal RNA (rRNA) in human, mouse, and rat samples. The amount of 18S rRNA target in the unknown samples is then scaled against a Control HeLa RNA standard curve for accurate quantification that fails using conventional techniques. In addition to the Power SYBR Green RNA-to-CT 1-Step Kit, the kit also includes Control HeLa Total RNA for standard curve preparation and an 18S rRNA Primer Mix, providing an assay range of 0.01 pg–1 ng total RNA.

Unique method eliminates the need to isolate RNA from cultured cells
The Cells-to-Ct method features a unique lysis reagent that lyses cells and protects RNA. DNA removal occurs simultaneously in the 5-minute lysis step to save time and improve ease of use. Lysis is then stopped by the addition of a “Stop Solution” that irreversibly binds to the lysis reagents.

The SYBR Green Fast Advanced Cells-to-CT Kit contains Cells-to-Ct Fast Advanced reverse transcription (RT) reagents for cDNA synthesis and PowerUp SYBR Green Master Mix for real-time PCR analysis. Primer sets are provided by the user.

Simple 7-minute sample preparation
The SYBR Green Fast Advanced Cells-to-CT Kit is designed for 10–100,000 cultured cells/sample. Cells are washed in PBS and lysed in lysis solution for five minutes at room temperature; DNase treatment can be performed simultaneously. Lysis is terminated at room temperature by a 2-minute incubation with Stop Solution. The lysates are now ready for reverse transcription or storage at –20°C for a short term or -80C for a long term.

Maximize sensitivity of detection
Unlike some competitor kits that limit the amount of lysate in the RT reaction to 10%, the SYBR Green Fast Advanced Cells-to-CT Kit can accommodate 45% of the total RT reaction volume as cell lysate. Additionally, cDNA can comprise up to 30% of the real-time PCR reaction volume. The large lysate volume in the optimized RT reaction, along with the large cDNA volume in the subsequent real-time PCR using the PowerUP SYBR Green Master Mix, lead to maximum sensitivity. The master mix amplifies the target precisely and accurately, enabling the detection of small quantities of target, such as transcripts expressed at low levels.

Reduced sample loss and transfer error
Because samples can be processed directly in culture wells (96 or 384 wells), sample handling and the potential for sample loss or transfer error are minimized, facilitating rapid, high-throughput processing. Unlike old-fashioned, multi-step RNA isolation protocols, no heating, washing, or centrifugation steps are required. The kit greatly simplifies a laborious 30–60 minute process and reduces it to 7 minutes.

SYBR™ Green I Nucleic Acid Gel Stain, 10,000X concentrate in DMSO (Invitrogen™)

SYBR® Green I Nucleic Acid Gel Stain is one of the most sensitive stains available for detecting double-stranded DNA (dsDNA) in agarose and polyacrylamide gels. Because SYBR® Green I has greater sensitivity for dsDNA, it is especially useful for assays where the presence of contaminating RNA or ssDNA might obscure results. With exceptionally low background fluorescence and spectral characteristics that closely match light sources and filter sets in existing instruments, SYBR® Green I stain is ideal for use with laser scanners. SYBR® Green I stain provides:

Sensitivity—at least four times greater than ethidium bromide for DNA in agarose gels
Ease of use—gels soaked in diluted stain can be visualized without desalting

Applied Biosystems has developed a comprehensive set of guidelines to ensure success when using sequence detection systems (SDS) reagents and instrumentation. These guidelines are simple and easy to follow. Many traditional variables, such as magnesium chloride concentration and the thermal cycling protocol itself, have been standardized, greatly reducing assay development time.

All SDS reagent kits contain a passive internal reference to normalize non-PCR-related fluorescence fluctuations. Normalizing with a passive internal reference minimizes well-to-well variability that result from a variety of causes, such as pipetting errors and sample evaporation.

Real-time quantitative PCR was first performed using the 5' fluorogenic nuclease assay, which uses a TaqMan® probe during the PCR amplification to provide added specificity. An additional chemistry is also available, using SYBR® Green I dye, that can provide real-time quantitative PCR information. Assays using the SYBR® Green 1 double-stranded DNA binding dye do not require a TaqMan® probe and provide additional experimental flexibility.

The incorporation of SYBR Green I dye into a real-time PCR reaction lets the user detect any double-stranded DNA generated during PCR. This provides great flexibility because no target specific probes are required, and yet both specific and non-specific products will generate signal. The use of the hot-start enzyme AmpliTaq Gold® DNA Polymerase in all SYBR Green I reagent kits yields the highest performance available by minimizing non-specific product formation. SYBR Green I dyes are ideal for use in target identification (screening assays), or when only a small number of reactions are required for a given assay.

SYBR Green II RNA gel stain is a sensitive nucleic acid gel stain that has bright fluorescence when bound to RNA and low background in gels, making it ideal for use with either formaldehyde/agarose or polyacrylamide gels using laser scanners or standard UV transilluminators.
