          Assessing Critical Quality Attributes and Structural Conformation of Biotherapeutics by HDX-MS

          Hydrogen deuterium exchange (HDX) mass spectrometry (MS) is a powerful tool for studying the dynamics of higher order structure of protein-based therapeutics. The rate of hydrogen-to-deuterium exchange within the amide hydrogen on the backbone of biotherapeutics provides solvent accessibility information and thus protein structure and conformation can be inferred.

          Jennifer Zhang received her Ph.D. from University of California at Davis in 2004, where she was focused on characterizing the structures of O-linked oligosaccharides by MALDI-FTMS. After graduation, she worked at Thermo Scientific as a LTQ FT/Orbitrap product specialist. She was focused on developing applications for LTQ FT and LTQ Orbitrap to demonstrate their unique features in protein analysis including both bottom-up and top-down approaches. She joined Genentech in 2006 and has been working on analytical method development and qualification/validation, protein therapeutic structural characterization, process development support, regulatory submission content including IND and BLA filings. At Genentech, she is a technical leader and expert on a variety of mass spectrometric techniques to support detailed structural characterization of therapeutic proteins. She developed and established the first fully-automated HDX-MS system in Genentech. She leads a number of scientists and Post-docs to develop and apply HDX-MS techniques to characterize the higher order structure of protein therapeutics in support of drug development.

          Terry Zhang joined ThermoFisher scientific in 2001. Since then Terry had worked in Proteomics Marketing, Chromatography, and Food Safety and Environmental Marketing Groups. Terry currently is working in Pharma and Biopharma Marketing Group, covering protein characterization techniques such as protein higher order structure elucidation by hydrogen deuterium exchange mass spectrometry. Terry had a B.S. degree in Chemistry and an M.S. degree in Analytical Chemistry.
