The transfer of an analytical HPLC method between different instruments in regulated routine laboratories can be challenging. Similar difficulties can be faced when a laboratory reproduces methods from pharmacopeia monographs, typically on instruments different from those used for the monograph publication. There are many instrumental parameters that can influence absolute and relative retention times, as well as peak shape and peak resolution, or even relative peak areas. When a pre-validated standard method is to be reproduced, there is only little freedom for adapting method parameters in order to simulate the behavior of another instrument. To overcome such challenges, a software controlled and thus fully traceable tuning of relevant instrument parameters can be very convenient and will also fulfill regulatory compliance requirements.

This webcast details out how new and fully compliant technologies can deliver matching chromatograms in method transfer between different HPLC instrument by addressing:

  • Programmed and step-less gradient dwell volume adaptation
  • Adaptation of column thermostatting and eluent pre-heating characteristics
  • Pre-column mixing of sample injection zones with surrounding eluent
  • Tuning of detector settings for consistent peak shapes and area ratios

Key learning objectives:

  • The challenges in method transfer and their theoretical background
  • How the deviations in the chromatograms between systems can be systematically related to a root course from the instrument specifics
  • How new innovative method transfer tools successfully work on typical EP and USP methods

Who should attend?

  • Technicians, scientists and managers in small molecule pharma laboratories in charge of HPLC analyses in a regulated environment

Marketing Manager of HPLC group | Thermo Fisher Scientific

Carsten Paul is a marketing manager in the HPLC group at Thermo Fisher Scientific, based in Germering, Germany. Carsten joined Thermo Fisher Scientific as Application Specialist in 2012 working on application for Charged Aerosol Detectors and various Thermo Scientific Vanquish HPLC and UHPLC instruments. Later he was involved in in different functions for the launches of all Thermo Scientific Vanquish HPLC and UHPLC systems, including the latest additions of Vanquish Duo and Vanquish Core. Carsten is by education an environmental chemist and holds a PhD from the Friedrich Schiller University of Jena in Analytical Chemistry. For this studies he used MS based metabolomic technologies to study the molecular interaction of unicellular algae.
