The three Dynabeads SARS-CoV-2 Spike ELISA kits detect and quantify human IgG, IgM, or total Igs from serum or plasma samples from SARS-CoV-2 positive individuals and can be used both for qualitative and quantitative assays.

Dynabeads in ELISA accelerates detection and quantification of human SARS-CoV-2

Rapid Binding Kinetics with Bead-based ELISA

Rapid binding kinetics are at work in the bead-based ELISA method due to the relatively short distance between beads in suspension and the antibody. This leads to a much shorter incubation time compared to a plate-based ELISA method where long incubation times are necessary to ensure that the antibody settles at the bottom of the well to find its antigen.

A new plates-to-beads concept

The conventional plate-based ELISA method is associated with lengthy protocols requiring many wash steps, long incubation times and a lot of hands-on work, leading to a total run time of at least 2–5 hours. Additionally, conventional ELISA plates are coated with immobilized molecules with the aim of capturing corresponding enzyme bound antigens, antibodies or proteins. After washing, the activity of the microplate well-bound enzyme is measured.

The plates-to-beads concept is designed with the user in mind, simplifying the ELISA protocol and removing the need for long incubations and countless manual interventions. This is achieved through combining the known Dynabeads technology and ELISA protocol, resulting in a bead-based ELISA.

The Dynabeads magnetic beads are perfectly spherical and free floating in suspension during the assay run. The proximity to the antibodies while in suspension and the binding capacity due to the large total surface area of the beads leads to a more efficient capture while greatly decreasing incubation time, regardless of whether you choose to automate your solution or not. This stands in stark contrast to traditional plate-based ELISA assays where long incubation times are necessary to ensure the antibody settles at the bottom of the well to find its antigen.

Protocols for Dynabeads SARS-CoV-2 Spike IgX ELISA kits

Automated protocol

The automated protocol is based on the KingFisher instruments, software and plastic. Both KingFisher Flex and KingFisher Apex take 24-well plates and 96-well plates. Please refer to the product guide for a thorough run-through the protocol.

  1. Resuspend Dynabeads magnetic beads thoroughly and dilute the wash buffers according to pack insert
  2. Reconstitute the antibody controls and create a serial dilution
  3. Aliquot the sample, the resuspended Dynabeads, antibodies, and buffers into KingFisher plates
  4. Start the protocol and add plates according to the instructions—press start and walk away
  5. Add the stop solution and transfer to an ELISA plate for absorbance readout
  6. Read plate at 450 nm

Manual protocol

A mixer/roller and a DynaMag magnet is required for the washing steps. Please refer to the product guide for a thorough run-through the protocol.

Precision

Operator pipetting, instrument performance, and little variation between critical reagent batches all contribute to the high assay precision. Intra-assay precision for IgG, Total Ig, and IgM in serum was performed by one operator with serum batch 1 and both bead verification batches. Inter-assay precision for IgG, Total Ig, and IgM in serum was calculated from n=9 experiments from three operators, over three different days, and three donors.

The sensitivity of the Dynabeads-based automated ELISA is presented as signal-to-noise according to the ICH Topic Q 2 (R1) guidelines.The signal-to-noise was determined by comparing measured signals from samples spiked with High (15,000 pg/mL), Medium (3,750 pg/mL), and Low (937.5 pg/mL) of IgG with those of blank samples.

The concentration in Low (green bars), corresponding to 937.5 pg/mL, are within the acceptable ratio of 2:1 (Limit of Detection, LOD) and can be reliably detected by the Dynabeads-based automated ELISA. The concentration in Medium (red bars), corresponding to 3,750 pg/mL are within the acceptable ratio of 10:1 (Limit of Quantification, LOQ) and can be reliably quantified by the Dynabeads-based automated ELISA. The quantification of IgG (pg/mL) from samples spiked with High, Medium, and Low amounts of IgG demonstrates that the recovery from the different concentrations are within the accepted industry standard (80–120%) (see table). Within this range, the sample matrix is considered to have minimal effect on the ability of the assay to accurately quantify the target.

The data is presented as a ratio between absorbance values from the sample over absorbance values obtained by the calibrator (Medium). For comparison, a plate-based method is included. A ratio of <1 shows that the target is absent while a ratio of > 1.3 indicates that the target is present. A ratio between 1 and 1.3 is indeterminate. In Qualitative—IgM graph, clinical Sample 3 and 9 are classified as IgM-positive when using the Dynabeads-based automated ELISA, while Clinical Sample 4 and 8 are identified as indeterminate. In contrast, in Qualitative—IgG graph, clinical samples 1, 3, 4, 5, 6, 7, and 8 are classified as IgG-positive using the same method.
