An LC-HRAM-MS method with Thermo Scientific TurboFlow online SPE was developed and validated for the quantitation of immunosuppressants. Online SPE efficiently extracts analytes while removing other components in a crude supernatant, significantly simplifying sample preparation. The use of full-scan data-dependent MS2 acquisition allows for high-resolution selected-ion monitoring quantitation of the analytes, confirming the analytes using MS2 fragmentation, as well as monitoring the signal intensities of major immunosuppressant metabolites.
