The HID NIMBUS Presto System is validated for use with the PrepFiler and PrepFiler BTA automated forensic DNA extraction kits according to the Scientific Working Group for DNA Analysis Methods (SWGDAM, December 2016).

HID NIMBUS Presto System purification process

The deck-mounted KingFisher Presto instrument is integrated on the automated liquid handling workstation and utilizes magnetic particle-based technology to facilitate high-quality nucleic acid purification. When coupled with PrepFiler or PrepFiler BTA extraction chemistry, DNA binds to the magnetic particles, cellular debris/inhibitors are removed, and the bound DNA is eluted through the movement of the magnetic particles across reagent plates (Figure 1).

A dual-position turntable allows processing of one plate while the integrated automated liquid handler simultaneously prepares and transports a second plate, without the need for manual intervention. The patented pipette tip technology creates a tight seal for superior liquid measurement accuracy and precision without tip distortion or aerosol generation during tip pick-up and ejection. Integrated tools and flexible format include barcode scanners to track samples and labware grippers to automatically transport samples around the deck.

Casework-type samples

A variety of known and non-probative casework-type samples were used to evaluate the ability of the HID NIMBUS Presto System to extract inhibitor-free DNA of the highest quantity and quality possible from common human identification sample types (Figure 3).

Accurate and contamination-free

A checkerboard pattern, alternating neat blood and saliva samples with extraction reagent blanks, was utilized to confirm that the HID NIMBUS Presto System does not introduce cross-contamination during automated extraction. The NIST F standard was used to assess the accuracy of the process (Figure 4).
