Gibco CTS OpTmizer Pro serum-free media formatted in three different sizes – BPCs, bottles, or vials

When scaling up a T cell manufacturing process for clinical and commercial manufacturing, it is important to select one that is specifically made for the cell type you are using. Used in >200 clinical trials and in commercialized therapies, the Gibco CTS OpTmizer family of serum-free media (SFM) is thoughtfully designed to meet the nutritional needs of high-density T cells, which is particularly important when scaling up. The result is that all our media can help maintain the desired central memory phenotype. In addition, whether you’re developing an autologous or allogeneic T cell therapy, CTS OpTmizer SFM are serum and xeno free and have extensive traceability documentation, including Drug Master Files and Regulatory Support Files.

What’s new: CTS OpTmizer SFM family now in 5 L and 10 L BPCs

To enable easier scale up, CTS OpTmizer basal media is available in 5 and 10 L bioprocess containers (BPCs). The supplement, 130 and 260 mL, is also available in BPCs.

The BPCs are Aegis5-14 film with C-Flex and polyvinyl chloride (PVC) lines to help enable scale-up to larger bioreactors more easily. A BPC with PVC tubing will help limit the number of open steps performed under an isolator or biosafety cabinet while the supplement is added to the base medium in addition to glutamine and cytokines. It also can enable a higher level of control and better risk management when it comes to media preparation and safety stocks.

CTS OpTmizer Pro SFM

Gibco CTS OpTmizer Pro SFM is a novel medium, developed for the growth and expansion of human T lymphocytes (e.g., CD4+, CD8+, polyclonal cells, or antigen specific T cells) used in allogeneic cell therapies. CTS OpTmizer Pro SFM improves central memory phenotype and cell growth by shifting the cell’s metabolism. The result is a larger population of central memory cells in a shorter period of time.

CTS OpTmizer Pro SFM come in Thermo Scientific Aegis5-14 Film, which allows compatibility with closed systems. Thermo Scientific Aegis5-14 Film is a five-layer cast film produced in a cGMP facility. The outer layer is a polyester elastomer coextruded with an ethyl vinyl alcohol (EVOH) barrier layer and an ultra-low-density polyethylene product contact layer. Aegis5-14 Film is manufactured using no animal derived components.

T cell proliferation in healthy donors evaluated using Gibco CTS OpTmizer Pro SFM

Figure 1: Enhanced cell growth was observed when cultured in the presence of CTS OpTmizer Pro SFM. Cells from six healthy donors were expanded in CTS OpTmizer Pro SFM. Cells were assessed at day 17. Improved growth that became more pronounced over time was observed as indicated by an average increase of 187% compared to Gibco CTS OpTmizer SFM, no phenol red.

CTS OpTmizer T Cell Expansion SFM, no phenol red

Gibco CTS OpTmizer T-Cell Expansion SFM, no phenol red is serum-free, xeno-free. Developed for the growth and expansion of human T lymphocytes for autologous therapies. This complete serum-free and xeno-free medium has demonstrated successful T cell expansion in cell culture plates up to large-scale bioreactors as effectively as serum-containing medium.
