High quality DNA for efficient cloning

To decrease screening effort, it is critical to use DNA fragments with low error rates in your high throughput cloning pipeline. GeneArt HTP Strings Fragments offer some of the lowest error rates on the market, helping improve your chances of picking the right clone the first time.

To demonstrate this advantage, a test set of 55 HTP Strings DNA fragments containing 49 heavy or light chain variable regions and 6 non-antibodies was assessed by next-generation sequencing and achieved an average of 95.1% correctness. To confirm the NGS results and demonstrate suitability of the product for the desired cloning application, 80 HTP Strings DNA Fragments (coding heavy and light chain variable regions) were cloned in appropriate vectors. For two clones of each sample, correctness was determined by PCR and sequencing. In total, 156/160 clones (97.5%) were correct. Moreover, at least one correct clone could be identified for all tested samples (100%).

Distribution of perfect molecules

Figure 1 is a bar graph of the distribution of perfect molecules, with 156 out of 160 clones showing the correct sequence.
