E-Gel Imager System FAQs

What components come with my system?

There are three full-system options to choose from: Each of these systems comes with the E-Gel Imager camera hood, camera hood power supply cable, camera hood USB 2.0 cable, the selected base with power cord (UV and blue-light bases) or DC connector cable (adaptor base), the E-Gel Universal filter, two CDs that include the E-Gel Imager Software package (GelCapture Imaging Software and GelQuant Express Analysis Software) one GelQuant Express software activation USB dongle, and a quick reference card.

Can I get other bases separately?

Yes, the UV Light Base, Blue-Light base, and the Adaptor Base can be ordered separately.

Can the UV light base be used for both ethidium bromide–stained and SYBR dye–stained gels?

Yes, when using both ethidium bromide–stained and SYBR dye–stained gels, we recommended using the UV Light Base with the Universal (orange) Filter for the ethidium bromide–stained gels and the green filter for the SYBR dye–stained gels. Use of the blue-light base for ethidium bromide–stained gels is not recommended, as the fluorescence signal emitted from the ethidium bromide–stained bands using a blue-light base is a tenth of the signal obtained with the UV light base.

Are there other filters available?

Yes, there are three filters available for use with this system. In addition to the Universal filter, the E-Gel Imager Qdot 625 filter and the E-Gel UV/SYBR Filter are available. All three filters can be purchased separately.

Does this system only work with E-Gel precast agarose gels?

No. This system is not only appropriate for imaging nucleic acid separation in E-Gel precast agarose gels, it’s also a perfect solution for documenting and analyzing nucleic acids in other agarose gels, such as NuPAGE Bis-Tris Gels and pour-your-own gels (agarose or polyacrylamide).

Can the E-Gel Imager System be used with proteins?

Yes, Using E-Gel Imager white light conversion screen, cat # 4473061, this accessory can convert the blue light emitted by the E-Gel Imager blue-light base, as well as the UV light emitted by the E-Gel Imager UV-light base, to white light. The E-Gel Imager white light conversion screen is compatible with protein gel stains such SimplyBlue Safe, SilverQuest Silver, SYPRO Ruby and Coomassie blue stains.

Does it have it to be Qdot 625 conjugate? What is the emission spectra range? Which light base do you recommend?

The E-Gel Imager System currently only works with proteins coupled to Qdot nanocrystals. Using the SpectraViewer tool on the Life Technologies website ( thermofisher.com/spectraviewer), we can see that the Ex/Em peaks for Qdot 625 conjugate are <300 nm and ~621 nm, respectively. Either the UV or the blue-light base can be used with the Qdot nanocrystals, but the UV base will provide slightly better signal intensity.

Is the E-Gel Imager compatible with protein ECL or AP detection systems?

The E-Gel Imager will not work with the ECL or AP detection systems.

How large is the E-Gel Imager System?

The E-Gel Imager System is a compact benchtop device that stands 43.7 cm in height (overall dimensions: 43.7 x 28.4 x 20.3 cm). It consists of the E-Gel Imager camera hood (35.6 cm in height) that fits on top of a base unit (11.9 cm in height).

E-Gel Imager Software FAQs

What are the two software programs that are used with the E-Gel Imager System?

GelCapture Imaging Software is used for the visualization and documentation of nucleic acid gels.
GelQuant Express Analysis Software is used for quantification analysis of nucleic acid gels.

What are the computer requirements to run the GelQuant Express Analysis Software?

Your computer will need to be running the Windows® XP professional or Windows® 7 professional 32-bit operating system in order to install and run the GelQuant Express software for gel capture and band quantification. Additionally, you will need to have administrative privileges on the computer.

Can I run the software on a Macintosh® computer?

GelQuant Express software requires the Windows® XP professional or Windows® 7 professional 32-bit operating system. It can run on Macintosh® computers, but only through one of these operating systems.

How do I install GelCapture software on the Windows® 7 Professional operating system?

Installing GelCapture software from the CD (Windows® 7 Professional)
  1. Launch the CD with the Internet Explorer® browser, right-click the GelCapture.exe installation file, and select the “Run as administrator” option.
  2. Select “Allow, I trust this program. I know where it’s from or I’ve used it before.” to allow the program to run.
  3. Follow the instructions provided by the installation wizard.
  4. Once installation is complete, click “Finish” to exit Setup.

What is the dongle used for?

The E-Gel Imager Quantitation Dongle, also known as the GelQuant Express software activation dongle, must be inserted into the computer in order to use GelQuant Express software. If the software dongle is not detected, the warning “Sentinel HASP key not found (H007)” will appear on screen.

What adjustments can be made to the image using the GelCapture software?

Exposure, sensitivity, and brightness can be adjusted using the GelCapture software. Captured images can then be exported to the GelQuant Express software for data analysis.

What are the two main analysis options for the GelQuant Express Analysis Software?

The 1D option is used for analyzing 1D gels, such as molecular weight determination. The Express option is designed for analysis of nucleic acids and protein gel images. In the Express mode, lane profiles are analyzed with the flexible adjustment of background values. Separate bands can be quantified as one entity for easier analysis of complex results (e.g., degraded or smear bands).

Does the GelQuant software in the E-Gel Imager System work for protein analysis such as quantification?

The GelQuant software can be used for quantifying protein gels. The image needs to be available in .jpeg or .tiff format. However, the E-Gel Imager is not recommended for the acquisition of protein gel images, which typically require white-light illumination.
