The Thermo Scientific Fluoroskan Microplate Fluorometer is a compact, robust fluorescence plate reader built to run variety of fluorometric and FRET assays with excellent performance. The Thermo Scientific Luminoskan Microplate Luminometer is a sensitive, DLReady certified luminescence plate reader built to perform superbly for a variety of luminescence applications, including flash, glow and BRET assays. The DLReady-certified Thermo Scientific Fluoroskan FL Microplate Fluorometer and Luminometer is a dual-mode versatile workhorse for fluorometric and luminometric assays. Fluoroskan, Fluoroskan FL and Luminoskan instruments are all equipped with powerful Thermo Scientific SkanIt Software, offering increased usability for setting up protocols and data analysis. Reagent dispensers: simultaneous dispense and read

Reagent dispensers enable easy and accurate reagent addition simultaneously as the instrument reads—a critical feature for flash-type reactions. All Fluoroskan and Luminoskan models are offered with three options of associated reagent dispensers: two dispensers, one dispenser, or no dispensers.

Flexibility to read 6- to 384-well plates

Fluoroskan, Fluoroskan FL and Luminoskan plate readers allow you to choose your sample format depending on your unique research and throughput needs.

High sensitivity for both top and bottom reading

Fiberless direct illumination optics for both top and bottom reading. Top reading is great for homogeneous, BRET and FRET assays, whereas bottom-reading becomes very important for non-homogeneous and cell-based assays. Bottom reading shortens the distance between the cell layer at the bottom of the well and the detector and minimizes any effect from the cell culture medium.

Dynamic range >6 decades

Reliably measure high and low signal samples with Fluoroskan and Luminoskan readers that provide both sensitivity and broad dynamic range in the same run.

Multipoint well reading

Read multiple locations in a single well by stipulating in SkanIt Software where in the well you want to read. This is especially important for non-homogeneous mixtures and cell-based assays where cells are not uniformly distributed in each microplate well. Multipoint measurements on Thermo Scientific fluorescence plate readers can also be used as an alternative or quantitative complement to imaging fluorospot assays.

Incubation and shaking

Incubate up to 45°C with orbital shaking.

Auto-calibration and self-diagnostics

Enables accurate fluorescence data with every run. If something isn't quite right, all three reliable fluorescence and luminescence plate readers can tell you before your run.

Highly sensitive DNA quantification

Fluoroskan Microplate Fluorometer, when used with the Invitrogen Quant-iT dsDNA high sensitivity assays, offers excellent sensitivity even in the lower DNA concentration range in the microplate format. This fluorescence plate reader detects as little as 0.2 ng of λ DNA (Figure 3), corroborating kit specifications.

Pharmacological readout is consistent with both Fluoroskan FL and Varioskan LUX plate readers

Staurosporine is a global kinase inhibitor; niclosamide induces apoptosis through a mitochondrial intrinsic pathway; and gambogic acid causes apoptosis induction by inhibiting proteasome function leading to apoptosis and cell death. In a standard dose-response experiment involving these three drugs, the pharmacological readout is the EC50, effective concentration 50, which is the concentration of the drug that gives half the maximal response. The maximum response of these drugs is complete cell death, so EC50 is the drug concentration at which half of the cells remained viable, as measured by Invitrogen alamarBlue Cell Viability Reagent. Regardless of which of our plate readers you use—the filter-based Fluoroskan or monochromator-based Varioskan LUX—the data agree (Figure 4).

A549 cells were plated in a 2D monolayer on Nunc cell culture plates, treated with varying concentrations of each drug for 24 hours. Viability was assayed using alamarBlue reagent and measured on two Thermo Scientific plate readers: the Varioskan LUX Multimode Microplate Reader and the Fluoroskan FL Microplate Fluorometer and Luminometer. These data demonstrate that the pharmacological readout is the same regardless of which reader is used.

All three drugs have a different EC50, but the two different microplate readers show the same EC50 for each drug. This data validates each reader’s accuracy in measuring and calculating pharmacological data (Table 1).

Flash-type ATP reactions with kinetic measurements

Get powerful luminescence detection of a flash-type ATP reaction performed by two of our reader models: the dedicated Luminoskan luminescence reader and the luminescence module of the dual-mode Fluoroskan FL microplate reader. Without access to the right equipment, researchers often rely on less-sensitive glow-type assays to detect ATP reactions. With simultaneous dispense-read technology, Luminoskan and Fluoroskan FL readers offer increased sensitivity with sub-attomole detection limits (Figure 5).
