SkanIt Software makes it easy to have just one or multiple Thermo Scientific microplate readers in your lab. You can choose to run all your instruments with SkanIt Software from a single computer or multiple computers, and since the software comes with unlimited installations, you can use any computer you like to perform data analysis from wherever you like.

Merge data

The ability to merge data is helpful in a variety of assay types and improves data visualization on graphs. This feature is especially useful with Fluoroskan and Luminoskan instruments when kinetic assays with dispensing are performed. You may find this feature particularly valuable if you:

  • Read baseline kinetically before reagent addition, dispense reagent to activate the reaction, and continue reading the kinetic signal.
  • Conduct assays with multiple reagent additions
  • Perform calcium flux measurements (Fluo3, Fluo4, Fura2).

What is SkanIt Cloud Library?

SkanIt Cloud Library is a collection of validated sessions—protocols that have analysis steps with real data and calculations—within Thermo Scientific SkanIt Software, which controls all Thermo Scientific microplate readers. Users can access this SkanIt Cloud Library for performing immediate measurements or to use the sessions as templates to create their own assay protocols. SkanIt Cloud Library is also a data source for training (including self-training) of new users of SkanIt Software. The library contains protocols and representative examples of the most frequently used optical detection technologies: absorbance, fluorescence, time-resolved-fluorescence, luminescence and AlphaScreen. Each SkanIt Cloud Library session has complete sets of measurements with the required calculations and graph processing steps from real assays. Instructions for running the assays are also provided. The library currently contains hundreds of protocols and continues to expand as new applications emerge.

How to use SkanIt Cloud Library?

SkanIt Cloud Library can always be accessed with the latest SkanIt version(s) when the PC is connected to Internet. The software is royalty free, so users of compatible Thermo Scientific microplate readers can freely install SkanIt Software on multiple PCs. Once SkanIt Software is installed, SkanIt Cloud Library is readily available for use. The Cloud Library icon is visible in the Home menu of the software (a), as well as in the Main menu (b), as shown below:

With the newest version of SkanIt Software, SkanIt Cloud Library has become available in a redesigned format, making it simpler to search protocols by instrument, detection technology, plate format or other relevant search terms (if you click to enlarge the image of the search tool, you see an example in which "Invitrogen" was used as a search term). After selecting the Cloud Library, the entire list of protocols appears and the specific term of interest can be typed in the search bar. The most common search terms are already listed just below the search bar—simply click for a faster search. Additional search terms, or tags, can be easily added by clicking the [ + ] button next to the search bar to quickly customize future searches. Popular Thermo Fisher Scientific kits such as Invitrogen PrestoBlue Cell Viability Reagent or Invitrogen Quant-iT dsDNA High-Sensitivity (HS) Assay Kit can also be easily located.

Easily search the extensive SkanIt Cloud Library for a protocol. Make use of an extensive and growing database of sessions containing notes with instructions for how to perform the assay, plate layout used, real assay data, measurement protocol, and all relevant calculations and graphs.

Once the selected cloud session is opened, general technical guidance for how to perform the assay can be found in the “Notes” section. Instructions on the “Plate Layout”, “Protocol” and “Results” sections are also included.

There are a couple of different ways to save your session, depending on your needs. If you do not need to change the protocol steps, click “Save” or “Save As” to keep the measurement data that was included in the Cloud Library session. Clicking “Save” or “Save As” makes the content of the session (except the protocol steps) editable as needed. If you need to change the protocol steps, click “Save As New” to enable that flexibility. When you click “Save As New”, the existing results data will be removed and all the content of the session will be editable as needed, including the protocol steps.

Save resources with FluoroSpot pre-screening feature of SkanIt software for microplate readers

For the first time ever, a microplate reader is able to give you an indication of the success of your FluoroSpot assay before you spend resources imaging the plate. This is a new technology offered by Thermo Scientific fluorescence microplate readers and made possible by the multipoint top-reading capability introducted with the 6th edition of SkanIt Software.

Often performed in cancer, infectious disease, and immunology research, a remarkable feature of the ELISpot assay is its sensitivity. High sensitivity is important in these fields because, for example, antigen-specific T cells are typically present at low frequencies in vivo. With ELISpot, it is possible to detect a single cytokine-producing cell from as many as one million bystander cells.

Detection of cell-secreted proteins can also be achieved using fluorescence-labeled detection reagents (including streptavidin fluorescent conjugates), and in this case the assay is known as FluoroSpot. ELISpot and FluoroSpot assays use the same protocol shown below for coating plates with capture antibodies, cell culture, and stimulation and washing steps. FluoroSpot has an advantage over ELISpot in that it is more suited for the simultaneous detection of multiple analytes, also known as multiplexing.
