Quality control and minimum yield guarantee

Our unique quality control processes help increase performance, speed, and capacity. The result is quick and efficient delivery. Below, find additional information about estimated yields based on length of oligo and starting synthesis scale, as well as minimum yields based on synthesize scale and purification option chosen.

When ordering custom oligos, remember that the scale of synthesis (25 nmol, 50 nmol, 200 nmol, 1 µmol, or 10 µmol) is the starting point for synthesis, not the guaranteed final amount. Oligonucleotide length, sequence, GC content, and coupling efficiencies can vary the yield of nmole amounts post-synthesis.
Note: We guarantee the total yield of oligonucleotide as a minimum number of OD units. Purification by HPLC, PAGE, and Cartridge will also affect final product yield.

Key features of our unique processes

We use a state-of-the-art automated process to increase performance, speed and capacity. This means you get quality Custom Primers quickly and efficiently. Custom DNA Oligos are available as standard deoxynucleotides, modified based, 5′ modified nucleotides (including a wide variety of fluorescent dyes and enzyme conjugates), and S-oligos for antisense studies. Five scales and four purity levels are available for DNA oligos in both tube and plate formats.

Our unique process includes 100% in-process trityl monitoring with additional LCMS and OD to verify yield. This comprehensive QC also applies to our Next-Day delivery service. Our oligo offering is complimented by easy online ordering and overnight shipping for rapid, reliable delivery. Consistent oligonucleotide quality is guaranteed and a comprehensive Certificate of Analysis accompanies each order, indicating quality, molecular weight, extinction coefficient, sequence, and melting temperature. No other oligo vendor provides superior performance or better value.

Additional features of our process

  • Final testing of statistically selected oligos by LCMS to further ensure the quality.
  • Certificate of Analysis includes oligo name; scale of synthesis; sequence and length; melting temperature (TM); amount in ODs, µg, and nmole; and molecular weight and extinction coefficient calculated from your exact sequence, including modification.
  • Label information on each vial is durably printed on clear transparent stock to allow full view of contents.
  • Fluorescent oligos shipped in amber tubes to protect light sensitive oligos.
  • Award winning Technical Services for complete workflow support.
  • Shipping in specially designed, easy-to-identify envelopes by express overnight delivery.

Estimated yields

Synthesis scales refer to the amount of starting material present, not the amount of final product produced. This is the same for all manufacturers of synthetic DNA. When a 50 nmole scale synthesis is specified, approximately 50 nmoles of the first base is added to the DNA synthesizer. For an average 25-mer, at least 25% of this starting material will result in failure sequences, hence it is not possible to produce 50 nmoles of full-length product from a 50 nmole scale synthesis. In addition, further losses occur during processing, transfer of material, and quality control.

Estimated nmole yield

The table below shows estimated nmole yield for desalted oligos at the various starting synthesis scales. This table converts the OD units into nmole values for an oligo with an equal and even distribution of each base. The actual nmol conversion from OD for a specific oligo may be different based on the composition and distribution of each base within the oligo.

Minimum yields

When ordering custom oligos, remember that the scale of synthesis (25 nmol, 50 nmol, 200 nmol, 1 µmol, or 10 µmol) is the starting point for synthesis, not the guaranteed final amount. Oligonucleotide length, sequence, GC content, and coupling efficiencies can vary the yield of nmole amounts post-synthesis. Note: We guarantee the total yield of oligonucleotide as a minimum number of OD units. Purification by HPLC, PAGE, and Cartridge will also affect final product yield.

The table below shows minimum guarantee yields (OD260). Please note that OD guarantee is valid for unmodified oligos 20 to 100 bases in length.

Yields for oligos under 20 bases are not guaranteed. In general they are about half of the values as shown in the table below.
