Cloud connectivity helps increase productivity

iBright Analysis Software is built on the Thermo Fisher Cloud web-based platform, and is part of the Thermo Fisher Connect suite. Data can be exported directly from iBright Imaging Systems and securely stored in Thermo Fisher Cloud. As the iBright Analysis Software is web-based, you can access, review, analyze, and share your data wherever an internet connection is available. Furthermore, with Thermo Fisher Connect you can determine instrument status, firmware edition, and usage history, providing an extra degree of control over the monitoring of your investment.

The image gallery can be sorted based on application, date, or file name, so your data are readily retrievable and your experiments stay organized. With 10 GB of Thermo Fisher Cloud storage (storage space is expandable), thousands of images can be collected into a portfolio of your projects. Image preview is adjustable, so you can scan through closely related data without opening each image individually (Figure 1).

Achieve faster results with auto-analysis

Image analysis, which can be complicated, is simplified to only a few clicks (Figure 3). Automatically analyze up to 4 mini blots or mini gels at a time from a single image for increased throughput. Powerful algorithms are designed to consistently and accurately select and identify lanes and band boundaries for molecular weight estimation, densitometry, or normalization (Figure 4). Real-time adjustments can be made to band detection sensitivity to fine-tune for low-expression proteins, and bands can be removed from analysis to eliminate background interference. Reports can be exported to PDF format, or the raw data can be exported to Excel software for further manipulation.

Figure 3. Manual analysis using iBright Analysis Software. Several HA-tagged proteins were expressed in HeLa cell extract using the Thermo Scientific 1-Step Human High-Yield Mini IVT Kit (Cat. No. 88890) and appropriate expression-ready clones. The resulting reaction mixtures were prepared for SDS-PAGE and electrophoresed. The proteins were transferred to a PVDF membrane using the Pierce Power Blotter (Cat. No. 22834), and the membrane was blocked and probed with the following primary antibodies: mouse anti-HA (Cat. No. 26183) and rabbit anti–cyclophilin B (Cat. No. PA1-027A). The membrane was washed and probed with the following secondary antibodies in TBS-Tween 20: goat anti-rabbit Alexa Fluor Plus 800 (Cat. No. A32735) and goat anti-mouse Alexa Fluor Plus 680 (Cat. No. A32729). The membrane was washed and imaged on the iBright FL1000 Imaging System. The expression levels of the proteins were quantified after western blotting using manual analysis (region mode) in iBright Analysis Software.

Figure 4. Automated analysis using iBright Analysis Software. PageRuler NIR protein markers (Cat. No. 26635; 1/20 dilution); 10 µL, iBright Prestained Protein Ladder (Cat. No. LC5615); 3 µL, and a dilution series of A431 lysate starting at 15 µg/lane were loaded on four gels and electrophoresed, and then all four gels were transferred simultaneously to separate nitrocellulose membranes, to test transfer consistency and efficiency. The proteins were transferred using the Pierce Power Blotter (Cat. No. 22834), and the membranes were blocked with Blocker FL (10X) Fluorescent Blocking Buffer (Cat. No. 37565) and probed with the following primary antibodies: Hsp90 rabbit PAb (Cat. No. PA3-013) and PDI mouse MAb (Cat. No. MA3-019). The membranes were washed and detected with the following secondary antibodies: DyLight 594–conjugated goat anti–rabbit IgG (Cat. No. 35561) and Alexa Fluor Plus 800 dye–conjugated goat anti–mouse IgG (Cat. No. A32730). The blots were then imaged on the FL1000 Imaging System and analyzed using the auto-analysis feature in iBright Analysis Software. The screen shows identification of the four separate blots as four independent analysis frames that can be edited and analyzed separately.

Bring it all together with a complete report

After analysis, view your images alongside your data report (Figure 5). Selection of data in the data table (blue highlight) highlights the corresponding band on the image (red highlight), so visual qualitative analysis is more fluid. Data can be filtered to view only desired parameters.
