The choice of whether to use one chemistry or another depends on the abundance of the target protein, the size of the protein, and the downstream application.

For separation of a broad range of proteins, two chemistries, Bis-Tris and Tris-glycine, have been optimized for performance and long shelf life. Select Bis-Tris gel chemistry for low abundance target proteins or when downstream applications require high protein integrity, such as posttranslational modification analysis, mass spectrometry, or sequencing. Our Invitrogen Bolt Bis-Tris Plus Mini Gels build on the legacy of the highly referenced Invitrogen NuPAGE Bis-Tris Gels and have a wedge-shaped sample well for loading up to 2X greater sample volumes.

To separate high-abundance proteins, select our robust Invitrogen Novex Tris-Glycine Mini Gels or Novex Tris-Glycine Plus Midi Gels, which offer maximum protein separation and crisp, straight bands. Tris-acetate gel chemistry, offered by Invitrogen NuPAGE Tris-Acetate Gels, is recommended for the separation of high molecular weight proteins up to 500 kDa. Novex Tricine Gels are available in a mini gel format and are designed for the separation of low molecular weight proteins and peptides.

Mini or midi gel tank?

The choice of mini of midi tank depends on your sample throughput needs. Invitrogen mini gels are available with up to 15 sample wells and Invitrogen midi gels are available with up to 26 sample wells. The Mini Gel Tank is compatible with all mini gels and can accommodate up to two gels per run. The streamlined, side-by-side tank design configuration enables convenient sample loading and enhanced viewing during use.

If you have higher throughput sample processing needs, the Invitrogen SureLock Tandem Midi Gel Tank is designed for easy and consistent vertical protein gel electrophoresis of 1 or 2 Invitrogen midi gels. The SureLock Tandem Midi Gel Tank is compatible with all NuPAGE and Novex midi gels and can accommodate up to two gels per run.

Bolt Bis-Tris Plus gels are precast polyacrylamide gels designed for optimal separation of broad molecular weights under denaturing conditions. Designed using neutral-pH formulation, the Bolt Bis-Tris Plus gel chemistry minimizes protein modifications or degradation that can result in poor band resolution.

The WedgeWell design allows detection of proteins in very dilute samples or visualization of low-abundance proteins.

Bolt gels are an excellent choice for routine protein separation needs or when the target protein is of low abundance and when preserving protein integrity is crucial as in posttranslational modification analysis, mass spectrometry, or sequencing. About Novex Tris-Glycine Gels:

The Tris-glycine gel chemistry is robust for a wide range of sample types. Based on the traditional Laemmli system, Novex Tris-Glycine gels provide high-quality performance and separation of a wide range of proteins into well-resolved bands and are compatible with samples containing detergents and high salt concentrations. The Novex Tris-Glycine mini gels, WedgeWell format, feature easier-to-load, larger-capacity wells that let you load up to 60 μL of sample.

Novex Tris-Glycine mini gels are offered in nine polyacrylamide concentrations: 6, 8, 10, 12, 14, and 16 percent, and 4–12%, 4–20%, and 8–16% gradient and multiple well formats. About NuPAGE Tris-Acetate Gels:

The Tris-Acetate gel chemistry enables optimal separation of high molecular weight proteins when used with an SDS running buffer. NuPAGE Tris-Acetate gels offer an environment of pH 8.1 that minimizes protein modifications and results in sharper bands. NuPAGE Tris-Acetate gels can also be run with Novex Tris-Glycine Native Running Buffer to resolve native proteins more effectively than a Tris-glycine gel system.

The NuPAGE Tris-Acetate gels are available in two polyacrylamide concentrations: 7 and 3–8% gradient and multiple well formats.

About Novex Tricine gels:

Tricine gel chemistry enables the optimum separation of low molecular weight proteins and peptides. Novex Tricine gels are high-resolution gels for peptide and low molecular weight protein analyses. The Novex Tricine gel system is a modification of the Tris-glycine system in which tricine replaces glycine in the running buffer. This system uses a discontinuous buffer system specifically designed for the resolution of low molecular weight proteins.

The Novex Tricine gels are offered in three polyacrylamide concentrations: 10, 16 and 10–20% gradient and multiple well formats.

Protein gels welcome packs for protein expression are available in mini gel and midi gel formats designed for use in protein expression experiments and subsequent purification steps. SimplyBlue SafeStain, included in the protein gels welcome packs for protein expression, is a ready-to-use, fast, sensitive, and safe Coomassie G-250 stain for visualizing protein bands on polyacrylamide gels. The SimplyBlue SafeStain protocol is easy to perform and can be completed in less than three hours. For added speed, a simple microwave procedure can be completed in 12 minutes. Destaining is not required but may be performed with deionized water to achieve maximum sensitivity. This stain is completely non-hazardous and does not require methanol or acetic acid fixatives or destains, creating a healthier lab environment.
