Extract total protein from mammalian tissue or cell samples using mild but efficient lysis reagents. These reagents have been optimized and validated with specific tissue types, as well as in primary and cultured mammalian cells. Protein extracts are compatible with most protein assays and typical downstream applications. These reagents offer higher protein yield than other commercially available or homemade formulations.

These convenient single formulation reagents typically allow samples to be processed in 5–10 minutes. When lysing primary and cultured cells, mechanical disruption is not required.

Cells were harvested at 85% confluency, washed twice and collected in ice-cold PBS and counted. For each cell type, 1 x 10^6 cells were pelleted by centrifugation at 2,000 x g for 5 minutes and lysed in 1 mL M-PER Reagent for 5 minutes. The cell lysates were clarified by centrifugation at 14,000 x g for 10 minutes and the supernatant was collected and the protein concentration (µg/million cells) was determined using the Pierce BCA Protein Assay (Part No. 23227).
