Easy and efficient
I speak to customers all over the world, and the one thing they all desire is a product that is easy to use and that works well. Molecular Probes immunocytochemistry and live imaging kits for stem cells are definitely among these products. In 45 minutes with 6–7 easy steps, these kits show you if your stem cell colonies express the classical self-renewal marker TRA-1-60. I like the versatility of the kits—they are designed with three separate fluorescent probes, at least one of which should match the cubes in your microscope. The kits also offer detection of the CD44 marker for those who culture their stem cells on mouse feeders.

Customers have been most excited to see the use of these kits during the reprogramming sequence. The kits allow staining of reprogrammed colonies at various time points during the process, providing confidence that the positively staining emerging colonies are on the right track. Another valuable feature of the kits is that, at the end of the reprogramming sequence, it is easy to choose positive colonies, pick and transfer each colony to a new well, and verify reprogramming success, as the colony still displays the fluorescent markers in the new well. The kits are also helpful in distinguishing between partially and fully reprogrammed cells.

Technical Highlights

Enable successful reprogramming of PBMCs to iPSCs

Human induced pluripotent stem cells (iPSCs) derived from adult somatic cells hold great promise for disease modeling and clinical therapies. Most iPSCs are derived from fibroblasts donated from skin biopsies and expanded prior to reprogramming. The ability to use blood from donors or sample banks of frozen peripheral blood mononuclear cells (PBMCs) is attractive, but reprogramming from peripheral blood sources has cytotoxicity and low reprogramming efficiency challenges. Our research goal was to enable successful and consistent generation of iPSCs from frozen PBMCs using the CytoTune–iPS 2.0 Sendai Reprogramming Kit, by optimizing spinoculation, polybrene, cytokine, and other workflow conditions.
