Is c-Met Activated in a Ligand-Independent Way in Tumor Oncogenesis?

  • ABfinity™ c-Met and EGFR rabbit recombinant monoclonal antibodies for sensitive detection
  • Enable consistent performance for your assays
  • Consistent product from lot to lot

ABfinity™ recombinant rabbit monoclonal antibodies are produced from specific recombinant clones so antibody performance is consistent over time. ABfinity™ antibodies are available for many targets, including c-Met and EGFR.

The c-Met receptor is a potential therapeutic target in some cancer models.
c-Met is activated by phosphorylation at tyrosine residues, and can be activated by epidermal growth factor receptor (EGFR) without production of hepatocyte growth factor (HGF), inducing signaling cascades through a ligand-independent event. ABfinity™ recombinant monoclonal antibodies are ideal reagents for signaling pathway research. The ABfinity™ technology helps ensure consistent antibody performance so you don’t have to reoptimize your assay with each new lot.

Invitrogen™ Luminex® Assays for the Met Signaling Pathway

  • Fast and easy assay protocols—perform and analyze your data in less than one day
  • Superior performance—accurate, reproducible, and sensitive quantification of signaling proteins
  • High quality—antibody-based assays with excellent specificity and sensitivity

To facilitate the analysis of STAT3 as well as a broad range of important signaling molecules, Life Technologies has developed a series of singleplex and multiplex immunoassays for the Luminex® platform. Samples containing targeted analytes bind to antibodies immobilized on color-coded polystyrene or magnetic beads.

The beads are then analyzed with the Luminex® 100™ or 200™ system or the FLEXMAP 3D® system. By monitoring the spectral properties of the beads and the amount of associated RPE fluorescence (from the secondary antibody conjugate),
the concentrations of individual analyte targets in both singleplex and multiplex assays can be determined simultaneously. This workflow helps save cost, time, and sample.

A member of the tyrosine kinase superfamily, c-Met is the receptor for hepatocyte growth factor, also known as scatter factor (HGF/SF). Phosphorylation of tyrosines 1234 and 1235 of c-Met is required for activation of the receptor’s tyrosine kinase activity. Several signaling proteins are phosphorylated and/or localized in response to c-Met phosphorylation. STAT3 (signal transducer and activator of transcription 3) is phosphorylated on tyrosine residue 705. STAT3 mediates the expression of a variety of genes in response to cell stimuli, and thus plays a key role in many cellular processes such as cell growth and apoptosis.

Interrogate c-Met Proteins for Cancer Research Using ELISA

  • Reliable—get quantitative results with validated kits
  • Convenient—ready-to-use kits that come with standards and strip-well plates
  • Fast—assay protocol in 4 hours

c-Met ELISA kits allow easy and quantifiable measurement of soluble c-Met or phosphorylated c-Met. c-Met is a receptor tyrosine kinase for hepatocyte growth factor (HGF). Soluble c-Met is a truncated form of the c-Met membrane receptor and can be measured in serum, plasma, or supernatant. Phosphorylated and total c-Met can be measured in cell lysate samples.

Receptor tyrosine kinases are important therapeutic inhibition targets for cancer research.

c-Met has been shown to be overexpressed in a number of malignancies. Ligand binding of HGF to c-Met receptor triggers phosphorylation of tyrosines 1234 and 1235, leading to a cascade of downstream events including proliferation, angiogenesis, migration, and metastasis.

The c-Met ELISA kits are valuable assay tools for studying the effects of drug inhibitors on c-Met signaling. For normalizing c-Met content of the samples, the c-Met (Total) ELISA Kit is also available, which assays for c-Met independent of phosphorylation status. The assay is completed using a 4-hour incubation protocol.

Treatment with sodium orthovanadate
Figure 3. Treatment with sodium orthovanadate, a tyrosine phosphatase inhibitor, increased phosphorylation of tyrosines at positions 1230, 1234, and 1235 of c-Met in A549 cells.
A549 cells were treated with sodium orthovandadate at 1 mM for 16 hours, lysed, and assayed in parallel for both phospho–c-Met  and total c-Met. The level of c-Met phosphorylation has been normalized to total c-Met content.

Gibco® Recombinant Human Hepatocyte Growth Factor (HGF)

  • High biological activity
  • High purity
  • Proven compatibility with Gibco® media

Gibco® recombinant proteins are highly purified bioactive proteins. Gibco® cytokines and receptors have been bioassayed with Gibco® media. Get your recombinant proteins from the media company you use and trust.

HGF (hepatocyte growth factor) is the ligand of the MET receptor. Met signaling has been shown to play a role in cancer, and is a target for potential therapeutic research. HGF stimulation results in invasive growth. HGF acts by regulating cell growth, cell motility, and morphogenesis.

We offer high-purity recombinant human HGF for use in cell culture applications. Every lot of HGF is tested for bioactivity by demonstrating the dose-dependent proliferation of monkey 4MBr-5 cells.
Gibco® cytokines, growth factors, and recombinant proteins are of high purity and high bioactivity. To ensure that Gibco® growth factors are of the highest quality, each protein is analyzed for purity along with structural homogeneity so that a biologically active protein is produced.
