Tips and Tricks

  • CELLstart™ substrate should be diluted in DPBS that contains calcium and magnesium  (Cat. No. 14040141) because these inorganic salts help facilitate binding. CELLstart™ substrate can be used with human embryonic stem cells (hESCs), mesenchymal stem cells, neural stem cells, and  human induced pluripotent stem cells (iPSCs), and for the attachment of human feeder cells (e.g., foreskin fibroblasts).

  • Geltrex™ Basement Membrane Matrix can be used for more than just a coating layer for your hESCs! Geltrex™ matrix can also be used for cell invasion assays by applying a thick layer— 50–200 μL per cm2 --onto the growth surface. Incubate your plate for 30 minutes, and you're ready to go!

  • Cryopreservation of hESCs using KnockOut™ SR XenoFree. Prepare cryopreservation medium by supplementing KnockOut™ SR XenoFree Complete Medium with an additional 10% KnockOut™ SR XenoFree (to yield a final concentration of 25%) and 10% dimethyl sulfoxide (DMSO) cryoprotectant. Expect some cell death at recovery, and freeze hESCs at a higher density than would normally be passaged. (If cells are routinely passaged at a 1:5 dilution, a 1:3 or 1:4 dilution is recommended). .) Gently resuspend the cell pellet with cryopreservation medium without triturating. While transferring the contents into vials, invert the capped hESC tube regularlyroutinely to mix the cells. For best results, hESC vials should be Nalgene Cryo 1°C Freezing Containers).

Troubleshooting/FAQs

  1.    Can human MSCs be transferred directly into StemPro® MSC SFM XenoFree medium from FBS-supplemented medium?

Yes. Our studies show that MSCs transferred directly into StemPro® MSC SFM XenoFree demonstrate comparable expansion anddoubling times as DMEM + 10% MSC-qualified FBS.

  2.    What does it mean to be "XenoFree"?

XenoFree (XF) Component (Direct)—a component will be considered to be XenoFree (direct) if it has not been directly derived from animal tissue, cells, or body fluid except for tissues, cells, or body fluids of human origin. Components produced via bacterial, yeast, or other recombinant processes will also be considered to be XenoFree origin even if the manufacturing process includes animal components in the fermentation broth or process enzymes
