Protein yield from various cell types using M-PER Mammalian Protein Extraction Reagent

Cells were harvested at 85% confluency, washed twice and collected in ice-cold PBS and counted. For each cell type, 1 x 10^6 cells were pelleted by centrifugation at 2,000 x g for 5 minutes and lysed in 1 mL M-PER Reagent for 5 minutes. The cell lysates were clarified by centrifugation at 14,000 x g for 10 minutes and the supernatant was collected and the protein concentration (µg/million cells) was determined using the Pierce BCA Protein Assay (Part No. 23227).
