Immunofluorescence analysis of CD49b/ITGA2 was performed using 70% confluent log phase U-2 OS cells. The cells were fixed with 4% paraformaldehyde for 10 minutes and blocked with 2% BSA for 1 hour at room temperature. Description: The eBioY418 (Y418) monoclonal antibody reacts with human CD49b, the 150-kDa, integrin alpha 2 subunit. The complex of CD49b non-covalently associated with integrin beta 1 (CD29), also known as VLA-2, is a receptor for collagen and laminin. This complex is expressed by lymphocytes, activated T cells, monocytes and platelets.
Applications Reported: This eBioY418 (Y418) antibody has been reported for use in flow cytometric analysis. (Fluorochrome-conjugated eBioY418 is recommended for use in flow cytometry.).
Applications Tested: This eBioY418 (Y418) antibody has been tested by flow cytometric analysis of normal human peripheral blood cells. This can be used at less than or equal to 0.25 µg per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
Purity: Greater than 90%, as determined by SDS-PAGE.
Aggregation: Less than 10%, as determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
CD49b (Integrin alpha 2, Integrin alpha 2 chain, VLA-2 alpha chain, HM alpha 2) is a member of the integrin family. It is a glycoprotein with molecular weight of 150 kD, and it complexes with CD29 (Integrin beta 1) to form the heterodimeric integrin VLA-2 (integrin alpha 2 beta 1, or GPIa/IIa) complex. VLA-2 is an extracellular receptor for laminin, collagen, and fibronectin, and interaction with its ligands results in the activation of intracellular signaling pathways. It has reported roles in VEGF-induced angiogenesis in vivo, as well as adhesion and lymphocyte activation. CD49b is expressed by NK cells, NK-T cells, monocytes, platelets, and epithelial cells. It is also expressed on adaptive immune cells such as T and B cells, specifically on a subset of CD4+ T cells in the spleen, on intraepithelial and lamina propria lymphocytes in the intestine, as well as on a population of peripheral CD4+ type 1 T regulatory (Tr1) cells that co-express LAG-3.
