Ipilimumab (Yervoy) is a humanized monoclonal antibody that is approved by the U.S. Food and Drug Administration for the treatment of patients with unresectable or metastatic melanoma. Ipilimumab works to activate the immune system by targeting CTLA-4 (Cytotoxic T-lymphocyte protein 4), a negative regulator of cytotoxic T-lymphocyte activity. Ipilimumab binds to CTLA-4, blocking the inhibitory signal, which allows the cytotoxic T-lymphocytes to destroy the cancer cells.Anti-Ipilimumab Antibody (4H6), mAb, Mouse is produced from a hybridoma resulting from the fusion of partner and B-lymphocytes obtained from a mouse immunized with Ipilimumab.
Upon reconstitution, it can be stored for 2-3 weeks at 2-8 °C or for up to 12 months at -20 °C or below.
CTLA4 (Cytotoxic T-Lymphocyte Antigen 4) is a CD28-family receptor expressed on mainly CD4+ T cells. It binds the same ligands as CD28 (CD80 and CD86 on B cells and dendritic cells), but with higher affinity than CD28. However, in contrast to CD28 which enhances cell function when bound at the same time as the T cell receptor, CTLA4 inhbits the T cell and prevents it from functioning. Intracellular CTLA4 is also found in regulatory T cells and may be important to their function.
