ADAMTS13 Antibody in Immunoprecipitation (IP) — **Detection of human ADAMTS13 by western blot and immunoprecipitation.** *Samples:* Pooled normal human serum (300 µl/IP, 10% of IP loaded; 1.5 µl for input). *Antibodies:* Agarose immobilized goat anti-ADAMTS13 antibodies S300-391, S301-000, S301-001, S301-002 and S301-003 were used at the equivalent of 35 µg of antibody per IP. For blotting immunoprecipitated ADAMTS13, goat anti-ADAMTS13 antibody A300-002A was used at 1 µg/ml. *Detection:* Chemiluminescence with an exposure time of 30 seconds. Note: The anti-ADAMTS13 antibodies A300-391A, A301-000A, A301-001A, A301-002A and A300-003A where conjugated to agarose to produce the agarose immobilized antibodies S300-391, S301-000, S301-001, S301-002 and S301-003, respectively.

Target Information

ADAMTS13 is a member of the ADAMTS (a disintegrin and metalloproteinase with thrombospondin motif) protein family. Members of the family share several distinct protein modules, including a propeptide region, a metalloproteinase domain, a disintegrin-like domain, and a thrombospondin type 1 (TS) motif. Individual members of this family differ in the number of C-terminal TS motifs, and some have unique C-terminal domains. The enzyme is the von Willebrand Factor (vWF)-cleaving protease, which is responsible for cleaving at the site of Tyr842-Met843 of the vWF molecule. A deficiency of this enzyme is associated with thrombotic thrombocytopenic purpura.
