PA1-1750 detects ADAMTS4 from porcine, bovine, and human samples. PA1-1750 is specific for ADAMTS4 and shows no cross-reactivity with other ADAMTS family members.
PA1-1750 has been successfully used in Western blot procedures. By Western blot, this antibody detects an ~68 and an~53 kDa protein representing ADAMTS4 from human recombinant ADAMTS4. Depending on the extent of C-terminal processing, several smaller bands may also be seen using this antibody.
PA1-1750 immunogen is a Synthetic peptide corresponding to residues C Y(590) N H R T D L F K S F P G P(603) of human ADAMTS4. This polyclonal antibody has been referred to as: JSCYNH.
A disintegrin-like and metalloproteinase thrombospondin type 1 motif 4 (ADAMTS4 or aggrecanase 1) is a member of the ADAMTS family of multidomain proteins that has demonstrated metalloproteinase functions. The ADAMTS4 protein contains a prohormone convertase cleavage site, thrombospondin-1 like domain, cysteine-rich domain, and a propeptide domain. ADAMTS4 activity had been shown to be blocked by TIMP-1, 2, 3, and 4.
ADAMTS4 plays an important role in the cleavage of aggrecan, versican and brevican. Studies have determined that aggrecan is cleaved within the interglobular domain of aggrecan at sites Glu(373)-Ala(374) and Phe(342)-Glu(373). Aggrecan degradation has been linked to the development of rheumatoid arthritis, osteoarthritis, retinal diseases, Alzheimer"e;s and other neurodegenerative diseases.

Interleukin-1alpha treatment of meniscal explants stimulates the production and release of aggrecanase-generated, GAG-substituted aggrecan products and also the release of pre-formed, aggrecanase-generated G1 and m-calpain-generated G1-G2.
PA1-1750 was used in western blot to investigate the effect of IL-1alpha on ADAMTS4 expression in the meniscal fibrocartilage

Activation of the proteolytic activity of ADAMTS4 (aggrecanase-1) by C-terminal truncation.
PA1-1750 was used in western blot to investigate the mechanism for the activation of ADAMTS4 proteolytic activity

Versican V1 proteolysis in human aorta in vivo occurs at the Glu441-Ala442 bond, a site that is cleaved by recombinant ADAMTS-1 and ADAMTS-4.
PA1-1750 was used in western blot to investigate the proteolytic processing of versican ADAMTS isoforms
