ALKBH5 Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence staining of ALKBH5 in U2OS cells. Cells were fixed with 4% PFA, permeabilzed with 0.1% Triton X-100 in PBS, blocked with 10% serum, and incubated with ALKBH5 Polyclonal Antibody (Product # PA5-117781, 1:200) at 4°C overnight. Then cells were stained with the Alexa Fluor®488-conjugated Goat Anti-rabbit IgG secondary antibody (green). Positive staining was localized to nucleus.

ALKBH5 Antibody in Western Blot (WB) — Western Blot using ALKBH5 Polyclonal Antibody (Product # PA5-117781) at 1:500 dilution. Lane A: 293 Whole Cell Lysate, Lane B: U-251 MG Whole Cell Lysate. Lysates/proteins at 30 μg per lane. Secondary antibody: Goat Anti-Rabbit IgG (H+L)/HRP at 1:10,000 dilution. Developed using the ECL technique. Performed under reducing conditions. Predicted band size: 44 kDa. Observed band size: 48 kDa.

ALKBH5 Antibody in Immunoprecipitation (IP) — ALKBH5 Immunoprecipitation using: Lane A: 0.5 mg 293 Whole Cell Lysate 4 µL with ALKBH5 Polyclonal Antibody (Product # PA5-117781) and 60 μg of Immunomagnetic beads Protein A/G. Primary antibody: ALKBH5 Polyclonal Antibody, at 1:100 dilution. Secondary antibody: Clean-Blot IP Detection Reagent (HRP) at 1:1,000dilution. Developed using the ECL technique. Performed under reducing conditions. Predicted band size: 44 kDa. Observed band size: 50 kDa.

Target Information

ALKBH5 (RNA demehtylase ALKBH5) is a dioxygenase that demethylates RNA by oxidative demethylation. Specifically, it demethylates N(6)-methyladenosine (m6A) RNA, the most prevalent internal modification of messenger RNA (mRNA) in higher eukaryotes. Demethylation of m6A mRNA affects mRNA processing and export. ALKBH5 can also demethylate N(6)-methyladenosine in single-stranded DNA (in vitro). It requires molecular oxygen, alpha-ketoglutarate and iron.
