Western blot detection of AP2 alpha in 20µg of lysates from: (Lane 1) HeLa nuclear extract, (Lane 2) HeLa, (Lane 3) 293, (Lane 4) A431 and (Lane 5) Jurkat whole cell lysates. In (Lanes 6-10 the antibody was preincubated with 1 µg/ml of the immunizing peptide which effectively blocks the specific reactivity of this antibody with AP2A. Samples were separated on SDS-PAGE, transferred onto nitrocellulose, and probed using an AP2 alpha polyclonal antibody (Product # PA1-31776) at a dilution of 1:500 followed by detection using a 1:5000 dilution of HRP-labeled Rabbit anti-Goat IgG for 1 hour at room temperature. Chemiluminescence was used for signal detection at a 60-sec exposure time.

The protein encoded by this gene is a transcription factor that binds the consensus sequence 5'-GCCNNNGGC-3'. The encoded protein functions as either a homodimer or as a heterodimer with similar family members. This protein activates the transcription of some genes while inhibiting the transcription of others. Defects in this gene are a cause of branchiooculofacial syndrome (BOFS). Three transcript variants encoding different isoforms have been found for this gene.
