ARHGEF7 Antibody in Immunohistochemistry (IHC) — Immunohistochemical staining of ARHGEF7 in human cerebral cortex using ARHGEF7 Polyclonal Antibody (Product # PA5-52097) shows weak to moderate cytoplasmic positivity in neurons.

ARHGEF7 Antibody in Western Blot (WB) — Western blot was performed using Anti-ARHGEF7 Polyclonal Antibody (Product # PA5-52097) and a 76 kDa band corresponding to ARHGEF7 was observed across cell lines and tissue tested. Whole cell extracts (30 µg lysate) of A-431 (Lane 1), Caco-2 (Lane 2), Daudi (Lane 3), A549 (Lane 4), HEK-293 (Lane 5), Hep G2 (Lane 6), SH-SY5Y (Lane 7) and tissue extract (30 µg lysate) of Mouse Lung (Lane 8) were electrophoresed using NuPAGE™ 10% Bis-Tris Protein Gel (Product # NP0302BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (0.4 µg/mL) and detected by chemiluminescence Goat Anti-Rabbit IgG Secondary Antibody, HRP conjugate (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005). An uncharacterized band at ~32 kDa was also observed in certain samples tested.

ARHGEF7 Antibody — Antibody specificity was demonstrated by siRNA mediated KD of target protein. A-431 cells were transfected with ARHGEF7 siRNA and loss of signal was observed in Western Blot using ARHGEF7 Polyclonal Antibody (Product # PA5-52097). {KD}

Target Information

The serine/threonine kinase, p21 activated kinase (PAK), is a downstream effector of the small GTPases Cdc42 and Rac. PAK associates with Nck, the p85 and p110 subunits of PI3-kinase and PIX (PAK-interacting exchange factor) in a focal complex. The binding of PIX is necessary for the localization and activation of PAK in the Cdc42 to Rac signaling pathway, and this binding occurs through the high affinity of the N-terminal SH3 domain of PIX for a conserved proline rich PAK sequence. PIX exists as two isoforms, alpha and beta, and both are highly expressed in heart, muscle and thymus tissues of human and rat. alphaPIX is phosphorylated via PDGF and EphB2 receptor signaling pathways or through association with PI3-kinase. The alphaPIX isoform predominantly acts as a guanine nucleotide exchange factor (GEF) on Rac, which may mediate lamellipodia formation.
