0.25 µg/well of total overexpressed Hordeum vulgare ASY 1 protein extracted the previous day from Rosetta 2 p Lys S cells with Bug Buster master mix and Complete EDTA free protease inhibitor, captured with Ni NTA, digested at 4°C overnight with Pro TEV protease to remove the affinity tag, and run through a second round of Ni NTA capture to remove the cleaved affinity tag. Store lyophilized/reconstituted at -20°C; once reconstituted make aliquots to avoid repeated freeze-thaw cycles. Please, remember to spin tubes briefly prior to opening them to avoid any losses that might occur from lyophilized material adhering to the cap or sides of the tubes.

Target Information
Asynapsis 1 (ASY1) encodes a meiosis-specific chromosome-axis protein that localizes along the axial/lateral elements of prophase I chromosomes in developing meiocytes, where it is positioned at the bases of chromatin loops and becomes enriched on unsynapsed regions to help coordinate homolog pairing and synaptonemal complex assembly. The ASY1 protein is a largely helical, coiled-coil-rich factor with an N-terminal HORMA domain that mediates regulated association with axis partners and supports checkpoint-competent chromosome organization; multiple transcript isoforms have been reported, consistent with modular regions involved in axis targeting and protein-protein interactions. Functionally, ASY1 is required for robust establishment of the meiotic chromosome axis, timely synapsis of homologs, and efficient progression of recombination toward crossover formation, and loss of ASY1 disrupts homolog alignment/synapsis, alters recombination outcomes, and compromises fertility by impairing faithful reductional segregation during meiosis.
