Axl Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Axl Polyclonal Antibody detects AXL protein at cell membrane in human lung cancer by immunohistochemical analysis. Sample: Paraffin-embedded human lung cancer. Green: Axl Polyclonal Antibody (Product # PA5-77875) diluted at 1:200. The signal was developed using goat anti-rabbit IgG antibody (Dylight488). Blue: Nuclear staining with Hoechst 33342. Antigen Retrieval: Citrate buffer, pH 6.0, 15 min.

Axl Antibody in Western Blot (WB) — Western Blot using Axl Polyclonal Antibody (Product # PA5-77875). Various whole cell extracts (30 µg) were separated by 5% SDS-PAGE, and the membrane was blotted with Axl Polyclonal Antibody (Product # PA5-77875) diluted at 1:1,000. The HRP-conjugated anti-rabbit IgG antibody was used to detect the primary antibody.

Axl Antibody in ELISA (ELISA) — An ELISA plate is coated with 50 µL of AXL recombinant protein at concentration of 4 µg/mL. The coated protein is detected with anti-Axl Polyclonal Antibody (Product # PA5-77875) at concentration ranged from 0.25 to 1,000 ng/mL.

Axl Antibody — Antibody specificity was demonstrated by shRNA mediated knockdown of target protein. HeLa cells were transfected with Axl shRNA and decrease in signal intensity was observed in Western blotting using Anti-Axl Polyclonal Antibody (Product # PA5-77875).

Target Information

Axl is a member of the TAM receptor family, which also includes also MerTK and TYRO3. Axl contains protein kinase activity and is activated by dimerization and auto-phosphorylation upon ligand binding. In response to its ligands, Protein S and GAS6, Axl activates anti-inflammatory pathways (through SOCS-1 and SOCS-3), which limits TLR and cytokine signaling. This results in dampened inflammatory responses in macrophages and DCs. TAM receptors are also involved in phagocytosis of apoptotic cells. Mice lacking all three TAM receptors have several degenerative phenotypes linked to inefficient removal of apoptotic cells and membranes (e.g., in the retina and the male reproductive tract) and develop a severe autoimmune phenotype akin to systemic lupus erythematosus, including the production of broad spectrum auto-antibodies. In addition, Axl may function as a putative entry receptor for filoviruses. Axl is also used to identify a specific subpopulation of human blood dendritic cells, also referred to as Siglec 6+/Axl+ dendritic cells. Cellular expression of Axl can be upregulated by TLR ligands, such as LPS or poly I:C. Soluble Axl is generated by proteolytic cleavage of the membrane form. Increased plasma levels may be indicative of inflammation and cancer.
