BCL6 Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis BCL6 was performed using 70% confluent log phase Raji (B-cell lineage) cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 1% BSA for 1 hour at room temperature. The cells were labeled with BCL6 Polyclonal Antibody (Product # PA5-14259) at 1:100 dilution in 0.1% BSA, incubated at 4 degree Celsius overnight and then labeled with Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A27034) at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with SlowFade® Gold Antifade Mountant with DAPI (Product # S36938). F-actin (Panel c: red) was stained with Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image showing nuclear localization. Panel e represents control cells with no primary antibody to assess background. Panel f represents negative control, MOLT-4 (T-cell lineage). The images were captured at 60X magnification.

BCL6 Antibody in Immunohistochemistry (IHC) — Immunohistochemical analysis of formalin-fixed, paraffin-embedded human lymph tissue using a Bcl-6 polyclonal antibody (Product # PA5-14259), followed by HRP-conjugated secondary antibody and DAB staining.

BCL6 Antibody in Western Blot (WB) — Western blot analysis using a Bcl-6 polyclonal antibody (Product # PA5-14259) in mouse cerebellum tissue lysates (35 µg per lane).

BCL6 Antibody in Flow Cytometry (Flow) — Flow cytometry analysis of Jurkat cells using a Bcl-6 polyclonal antibody (Product # PA5-14259) (bottom) compared to a negative control cell (top) at a dilution of 1:10-50, followed by a FITC-conjugated goat anti-rabbit antibody

BCL6 Antibody — Antibody specificity was demonstrated by detection of differential basal expression of the target across cell models owing to their inherent genetic constitution. Target Information

Bcl-6 proto-oncogene product (a Kruppel-type zinc-finger protein) is mainly expressed in normal germinal center B cells and related lymphomas. Bcl-6 is involved in chromosome rearrangements at 3q27 in non-Hodgkin's lymphomas and Bcl-6 rearrangements have been detected in 33%-45% of diffuse large B cell lymphomas. Bcl-6 has been detected immunohistochemically in follicular lymphomas, diffuse large B cell lymphomas, Burkitt's lymphomas and in nodular, lymphocyte predominant Hodgkin's disease.
