Western blot analysis of BSA was performed by loading an unrelated protein (lane 1), 1ug (lane 2), 5ug (lane 3) and 10ug of BSA (lane 4) in reducing sample buffer and protein ladder onto an SDS-PAGE gel. Proteins were transferred to nitrocellulose membrane. Membrane was blocked in 2% skim milk in PBS. BSA was detected at approximately 69 kDa using a BSA polyclonal antibody (Product # A11133) at a dilution of 1:1000 in 1% skim milk in PBST overnight at 4 degrees, followed by a 1:2000 dilution of HRP-conjugated goat anti-rabbit IgG antibody at room temperature for one hour. Albumin is a soluble, monomeric protein which comprises about one-half of the blood serum protein. Albumin functions primarily as a carrier protein for steroids, fatty acids, and thyroid hormones and plays a role in stabilizing extracellular fluid volume. Albumin is a globular unglycosylated serum protein of molecular weight 65,000. Albumin is synthesized in the liver as preproalbumin which has an N-terminal peptide that is removed before the nascent protein is released from the rough endoplasmic reticulum. The product, proalbumin, is in turn cleaved in the Golgi vesicles to produce the secreted albumin.
