          CCL3 Antibody (PA5-47000) in IHC (P)

          Immunohistochemical analysis of CCL3 in immersion fixed paraffin-embedded sections of human tonsil. Samples were incubated in CCL3 polyclonal antibody (Product # PA5-47000) using a dilution of 10 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown) and counterstained with hematoxylin (blue).           Product Specific Information

          In sandwich ELISAs, less than 0.05% cross-reactivity with recombinant human (rh) MIP-1 beta, rhMIP-1 gamma, rhMIP-3 alpha, rhMIP-3 beta, and recombinant mouse (rm) MIP-1 alpha is observed. This antibody neutralizes the biological activity of recombinant human CCL3/MIP-1 alpha. In the chemotaxis assay this antibody will also partially neutralize the biological activity of rhMIP-1 beta at a 20 fold higher IgG concentration. This antibody will not neutralize the biological activity of rmMIP-1 beta or rmMIP-1 alpha.

          Reconstitute at 0.2 mg/mL in sterile PBS.

          Endoxin level is <0.10 EU per 1 µg of the antibody by the LAL method.

          Target Information

          Both MIP-1alpha and MIP-1beta are structurally and functionally related CC chemokines. They participate in host response to invading bacterial, viral, parasite and fungal pathogens by regulating the trafficking and activation state of selected subgroups of inflammatory cells (e.g. macrophages, lymphocytes and NK cells). While both MIP-1alpha and MIP-1beta exert similar effects on monocytes, their effect on lymphocytes differ; with MIP-1alpha selectively attracting CD8+ lymphocytes, and MIP-1beta selectively attracting CD4+ lymphocytes. Additionally, MIP-1alpha and MIP-1beta have also been shown to be potent chemoattractants for B cells, eosinophils and dendritic cells. Both human and murine MIP-1alpha and MIP-1beta are active on human and murine hematopoietic cells.
